Determination of glycine and threonine in topical dermatological preparations

In the present study, a single HPLC method was developed for the determination of glycine and threonine in cicatrizants. Two different preparations Of a Cream and an ointment, and the corresponding bandages, onto which the formulations were applied, were Studied. The method involved Matrix solubilisationn with dichloromethane, liquid-liquid isolation of gly and thr with aqueous 1N NaOH, and derivatization with phenylisothiocyanate. Reversed-phase HPLC separation was carried Out by gradient elution with 20mM aqueous NaClO4 and acetonitrile (from 90% to 30% aqueous NaClO4 in 10min) on a LiChrospher (R) 100 RP-18 cartridge (125mm x 4.6mm). Analytes were determined with a UV detector set at 245nm. Quantitation was accomplished by internal standardization with methionine. Linearity was studied in the range 60-120% of the concentrations expected for gly and thr (viz. for gly from 200 to 400 mu g ml(-1), and for thr from 100 to 200 mu g ml(-1)). In reference aqueous samples, linear correlation (r)was better than 0.99 for gly and thr, while in spiked matrix samples r ranged from 0.97 to 0.98. Recoveries were in the 95-105% interval, and precision (CV%, N=6) was better than 5% for both analytes either in cream, ointment or bandages. Microwave-Assisted Extraction and HPLC-UV-CD Determination of (S)-usnic Acid in Cladonia foliacea

Summary: A rapid HPLC-UV/PAD-CD protocol was reported for the analysis and identification of secondary metabolites in C. foliacea extract, with the most abundant analyte identified as (S)-UA. A versatile microwave-assisted extractive (MAE) protocol was established to quantitatively recover (S)-UA from the plant, achieving 420 mg from 100 g of dried C. foliacea. This extraction method proved to be a suitable protocol for producing (S)-UA from C. foliacea for biological, pharmaceutical, or commercial purposes.

The method achieved quantitative recovery with satisfactory clean-up, demonstrating high selectivity, sensitivity, and low detection limits in the ng mL(-1) range.

Summary: This study aimed to optimize the production process of mesenchymal stem cell-derived secretome, lyosecretome, by investigating sources of variability and modifying the manufacturing process. Different excipient ratios did not significantly affect the product quality, but increasing the total concentration of excipients improved the preservation of proteins and lipids. By concentrating the final product to achieve a specific protein concentration, the manufacturing process was standardized, resulting in a more reproducible composition and higher biological activity of the product.

Characterization and Separation of Platinum-Based Antineoplastic Drugs by Zwitterionic Hydrophilic Interaction Liquid Chromatography (HILIC)-Tandem Mass Spectrometry, and Its Application in Surface Wipe Sampling

Summary: In this study, a wipe sampling method combined with zwitterionic hydrophilic interaction liquid chromatography (HILIC-Z)-tandem mass spectrometry (MS/MS) analysis was developed for the simultaneous evaluation of oxaliplatin, cisplatin, and carboplatin surface contaminations. The experiment design approach was used to optimize chromatographic conditions, resulting in quantification limits ranging from 2 to 5 ng/mL for oxaliplatin and carboplatin, and between 170 and 240 ng/mL for cisplatin. The wipe desorption procedure is comparable to other AD sampling methods, enabling efficient sample preparation with an LC-MS/MS analysis time of less than 7 minutes.

Summary: In this study, poly lactic-co-glycolic acid (PLGA) based nanoparticles (NPs) were proposed as carriers for indocyanine green (ICG), and fluorescence quenching experiments were conducted by studying the interaction between ICG and chitosan oleate (CS-OA). The results showed that associating ICG with NPs can reduce cell viability after laser irradiation, opening up new possibilities for utilizing PDT mechanism in cancer therapy.

A New Perspective on SPME and SPME Arrow: Formaldehyde Determination by On-Sample Derivatization Coupled with Multiple and Cooling-Assisted Extractions

Summary: Formaldehyde (FA) is a toxic and carcinogenic compound, making it important to regulate FA-releasing substances in various industries. This study proposes the use of solid-phase microextraction (SPME) with O-(2,3,4,5,6-pentafluorobenzyl)-hydroxylamine (PFBHA) derivatization and gas chromatography for FA monitoring. It evaluates the use of polydimethylsiloxane (PDMS) as a sorbent phase combined with multiple SPME (MSPME) and cooling-assisted SPME methods for FA determination. The method shows satisfactory sensitivity, linearity, precision, and accuracy for FA detection in real-life samples.

Combining DoE and MASE: a winning strategy for the isolation of natural bioactive compounds from plant materials

Summary: The NADD program, while successful, is still considered a secondary approach. Biomass extraction is a critical step in the process and often requires large amounts of organic solvents and time. Optimizing these procedures can reduce costs and improve waste management. Microwave assisted solvent extraction (MASE) is a commonly used technique in drug discovery from natural sources. However, the one-factor-at-a-time (OFAT) approach limits the advantages of MASE. Combining MASE with DoE can make it more environmentally friendly, though the application of this combination is limited.

A strategy takes Yiqing tablets as an example to carry out simpler multi-component quantification and use fingerprint technology for comprehensive quality consistency evaluation

Summary: In this study, the quality of Yiqing tablet was comprehensively evaluated using multi-component quantification, multi-dimensional fingerprint construction, and antioxidant activity analysis. The results showed that the sample had 25 components with antioxidant activity according to the HPLC fingerprint and most bands of FTIR also showed antioxidant activity. Based on the evaluation results of HPLC and FTIR fingerprint, all samples were classified as first level, demonstrating the consistency of sample quality. This study provides a new idea for achieving comprehensive quality evaluation of traditional Chinese medicines.

Green sample preparation of medicinal herbs in closed digester block for elemental determination by ICP OES

Summary: This work presents a simple, cost-effective, and environmentally friendly method for determining the content of multiple elements in medicinal herbs using ICP OES. The optimized experimental parameters result in low dissolved organic carbon content and residual acidity concentration. The accuracy of the method is confirmed through the analysis of certified reference materials, and its environmental friendliness is validated using the AGREE metric. Principal component analysis is used to classify the medicinal herbs based on their inorganic constituent concentrations.

Separation and characterization of related substances of Lurasidone hydrochloride by LC-QTOF-MS techniques

The study emphasizes the importance of monitoring and controlling related substances in the manufacturing processes to optimize the process and ensure quality control.

Summary: This study investigated the cellular uptake of low, medium and high molecular weight polyethylene glycol (PEG) in MCF-7 cells using UPLC-MS/MS and CID techniques. The results showed that PEG600, PEG2K, PEG4K and PEG10K exhibited low toxicity and had limited cellular entry at concentrations of 5-1200 μg/mL. Additionally, the environmental friendliness of the developed analytical methods was evaluated.
