A simple, non-destructive, methodology based on FT-Raman spectroscopy was developed for the quantitative analysis of risperidone in commercially available film-coated tablets. A simple linear regression model was constructed based on standard tablets, prepared using the same manufacturing process as the commercially available. The tablets contained 0.27, 0.54, 1.08, 1.62, 2.16, 3.24 and 4.32 wt% risperidone. The most prominent Raman vibration of the active pharmaceutical ingredient at 1533 cm(-1), recorded using a home-made rotating system, was plotted against concentration. The model was tested on commercial film-coated tablets. The results were compared against those obtained by application of HPLC on the same samples. (C) 2008 Elsevier B.V. All rights reserved.

Comparative Study of Sample Carriers for the Identification of Volatile Compounds in Biological Fluids Using Raman Spectroscopy

Panagiota Papaspyridakou, Michail Lykouras, Christos Kontoyannis, Malvina Orkoula

Summary: Vibrational spectroscopic techniques, particularly Raman spectroscopy, are gradually replacing chromatographic methods in the identification of ethanol and other volatile substances in body fluids. This study proposes three carriers and describes their corresponding sample preparation methods for determining ethanol in human urine samples. Among the three carriers, the last one achieves a quick, simple, and inexpensive identification of ethanol with prevention of evaporation.

Formation and Characterisation of Posaconazole Hydrate Form

Michail Lykouras, Malvina Orkoula, Christos Kontoyannis

Summary: This study found that the polymorphic conversion of posaconazole from Form I to Form-S, which may affect bioavailability, is due to an interaction with water. However, the poor wettability of posaconazole Form I makes it challenging to completely wet the particles and produce pure Form-S. Therefore, for isolation, Form I should be dispersed in water and subjected to sonication for at least 10 minutes. Pure posaconazole Form-S was characterized as a hydrate form containing three water molecules per API molecule using X-ray powder diffraction (XRPD), Raman spectroscopy, attenuated total reflection (ATR) spectroscopy, thermogravimetric analysis (TGA), and optical microscopy.

Non-Destructive and Non-Invasive Measurement of Ethanol and Toxic Alcohol Strengths in Beverages and Spirits Using Portable Raman Spectroscopy

Panagiota Papaspyridakou, Panagiota Giannoutsou, Malvina G. Orkoula

Summary: The measurement of ethanol and toxic alcohol strengths in beverages and spirits is crucial for health reasons and for identifying adulterated products. Chromatographic and spectroscopic techniques have been used, but their application is time-consuming or requires sample pretreatment. In this study, Raman spectroscopy was used for fast and non-destructive measurements. A through the container method was developed for a non-invasive analysis. The method was validated with calibration curves and used to study the concentrations of alcohol in various products.

Quantitative determination of alcohols in human biological fluids through Raman spectroscopy: An alternative alcohol test

Panagiota Papaspyridakou, Michail Lykouras, Malvina Orkoula

Summary: “A novel method for the detection and quantification of ethanol and methanol in human urine, saliva, and blood serum using Raman spectroscopy was developed. Calibration curves were created for each alcohol-biological fluid system to determine the detection limits. Methanol had lower detection limits, but were still below intoxication levels, while ethanol had detectable concentrations below permissible limits. Saliva showed the lowest detection limits for both alcohols.”

Xiaoling Liu, Ting Yang, Lu Chen, Lili Lan, Guoxiang Sun, Ping Guo

Summary: In this study, the quality of Yiqing tablet was comprehensively evaluated using multi-component quantification, multi-dimensional fingerprint construction, and antioxidant activity analysis. The results showed that the sample had 25 components with antioxidant activity according to the HPLC fingerprint and most bands of FTIR also showed antioxidant activity. Based on the evaluation results of HPLC and FTIR fingerprint, all samples were classified as first level, demonstrating the consistency of sample quality. This study provides a new idea for achieving comprehensive quality evaluation of traditional Chinese medicines.
