Determination of chlorophylls and their derivatives in Gynostemma pentaphyllum Makino by liquid chromatography-mass spectrometry

The objectives of this study were to develop a high performance liquid chromatography-mass spectrometry (HPLC-MS) method for determination of chlorophylls and their derivatives in Gynostemma pentaphyllum Makino, a traditional Chinese herb possessing vital biological activities. Chlorophylls were extracted with a quaternary solvent system of hexane-acetone-ethanol-toluene (10:7:6:7, v/v/v/v), followed by separation of a total of 15 chlorophylls and their derivatives within 32 min using a gradient mobile phase of acetone, acetonitrile and methanol and a HyPURITY C18 column, with detection at 660 nm and flow rate at 1 mL/min. Identification was carried out on the basis of retention behavior, absorption spectra and mass spectra using atmospheric pressure chemical ionization (APCI) in positive ion mode for detection. Of the 15 analytes, chlorophyll a, chlorophyll b, pheophytin a and pheophytin b were quantified by using standard calibration curves, with the other 11 being quantified with an internal standard Fast Green FCF. Chlorophyll extracts in G.pentaphyllum were found to contain pheophytin a (2508.3 mu g/g), pheophytin a' (111.2 mu g/g), chlorophyll a (113.8 mu g/g), chlorophyll a' (11.0 mu g/g), hydroxypheophytin a (88.6 mu g/g), hydroxypheophytin a'(66.5 mu g/g), pyropheophytin a (76.0 mu g/g), hydroxychlorophyll a (23.8 mu g/g), pheophytin b (319.6 mu g/g), pheophytin b, (13.2 mu g/g), chlorophyll b (287.9 mu g/g), chlorophyll b' (11.1 mu g/g), hydroxychlorophyll b (15.0 mu g/g), hydroxypheophytin b (11.2 mu g/g) and hydroxypheophytin b'(8.5 mu g/g). Analysis of Furan and Its Derivatives in Food Matrices Using Solid Phase Extraction Coupled with Gas Chromatography-Tandem Mass Spectrometry

Summary: The objective of this study was to develop a method for simultaneous analysis of furan and its 10 derivatives in various food commodities. The study successfully separated furan and its derivatives using a HP-5MS column and GC-MS/MS within 9.5 minutes. The method could also distinguish different furan isomers. The method validation demonstrated that the developed method was reliable for the analysis of furan and its derivatives in commercial foods.

Prostate-specific antigen density and preoperative MRI findings as predictors of biochemical recurrence in high-risk and very high-risk prostate cancer

Summary: Patients with high-risk and very high-risk prostate cancer who undergo prostatectomy have a high risk of biochemical recurrence (BCR). This retrospective observational study aimed to identify the risk factors for BCR after radical prostatectomy in these patients. The study found that patients in the very high-risk group had a lower BCR-free survival rate and even if their PSA levels were undetectable initially after surgery, they still had a high risk of BCR. Preoperative MRI imaging stage and PSA density were found to be predictors of short-term BCR after prostatectomy.

Anti-Atopic Dermatitis Activity of Epi-Oxyzoanthamine Isolated from Zoanthid

Summary: Atopic dermatitis (AD, eczema) is a common skin condition that primarily affects children and adults, but current treatments have limited effectiveness and side effects. This study investigated the potential use of epi-oxyzoanthamine, a bioactive compound derived from Formosan zoanthid, for treating AD. In vitro and in vivo experiments showed that epi-oxyzoanthamine inhibited cytokine expression and reduced skin inflammation, indicating its potential as an alternative treatment for AD. Overall, the study highlights the importance of exploring new therapies for atopic dermatitis.

Effects of Oil and Processing Conditions on Formation of Heterocyclic Amines and Polycyclic Aromatic Hydrocarbons in Pork Fiber

Summary: This study aimed to monitor the formation of toxic compounds HAs and PAHs in fried pork fiber, a common meat product in Taiwan. The results showed that frying in sesame oil or lard produced more HAs in pork fiber processed at 160°C for 15 min compared to 200°C for 6 min. Sesame oil frying also resulted in higher levels of HAs than lard frying under the same heating conditions. Among the various HAs, Harman and Norharman were the most abundant. Precursors such as reducing sugar, amino acid, and creatine/creatinine played a vital role in HAs formation. For total PAHs, the highest level was observed in pork fiber fried in lard at 200°C for 6 min. Frying in sesame oil also resulted in higher levels of total PAHs compared to lard frying. Notably, the highly toxic benzo[a]pyrene was undetected in fried pork fiber. The precursor benzaldehyde played a more important role in PAH formation. Principal component analysis revealed that the formation mechanisms of HAs and PAHs in fried pork fiber were different.

Recent advances in the analysis of polycyclic aromatic hydrocarbons in food and water

Summary: This article provides an overview of recent advances in chromatographic and sensor-based analytical methods for the analysis of PAHs in food and water. It covers conventional sample preparation methods, advanced extraction techniques, gas chromatography coupled with various detection techniques, high performance liquid chromatography coupled with fluorescence, diode array or mass/tandem mass detection, and emerging supercritical fluid chromatographic techniques. It also discusses the development of electrochemical sensors and SERS-based optical sensors for onsite detection of PAHs. The review article serves as a research update and identifies research gaps for future studies.

Prophylactic Effects of Purple Shoot Green Tea on Cytokine Immunomodulation through Scavenging Free Radicals and NO in LPS-Stimulated Macrophages

Purple shoot green tea, in particular, exhibited strong antioxidant properties. Tea extracts were able to reduce inflammation and modulate immune cell responses by regulating cytokine expression.

A strategy takes Yiqing tablets as an example to carry out simpler multi-component quantification and use fingerprint technology for comprehensive quality consistency evaluation

Summary: In this study, the quality of Yiqing tablet was comprehensively evaluated using multi-component quantification, multi-dimensional fingerprint construction, and antioxidant activity analysis. The results showed that the sample had 25 components with antioxidant activity according to the HPLC fingerprint and most bands of FTIR also showed antioxidant activity. Based on the evaluation results of HPLC and FTIR fingerprint, all samples were classified as first level, demonstrating the consistency of sample quality. This study provides a new idea for achieving comprehensive quality evaluation of traditional Chinese medicines.

Green sample preparation of medicinal herbs in closed digester block for elemental determination by ICP OES

Summary: This work presents a simple, cost-effective, and environmentally friendly method for determining the content of multiple elements in medicinal herbs using ICP OES. The optimized experimental parameters result in low dissolved organic carbon content and residual acidity concentration. The accuracy of the method is confirmed through the analysis of certified reference materials, and its environmental friendliness is validated using the AGREE metric. Principal component analysis is used to classify the medicinal herbs based on their inorganic constituent concentrations.

Separation and characterization of related substances of Lurasidone hydrochloride by LC-QTOF-MS techniques
