Human Neutrophil Defensins and Their Effect on Epithelial Cells

Background: The aims of the present study include: 1) to localize human neutrophil defensin-1 (HNP-1) through HNP-3 in gingiva and in neutrophil extract-treated epithelial cell monolayers; 2) to determine the effects of HNP-1 on the epithelial cell viability, attachment, and spreading; and 3) to analyze the effect of HNP-1 on the bacterial adherence to epithelial cells. Methods: For localization of HNP-1 through HNP-3 in gingival tissue and in preincubated cell monolayers, immunohistochemical and immunocytochemical methods were used. Viability and proliferation of epithelial cells were determined with commercial kits after incubating the keratinocytes with different HNP-1 concentrations (low, 1 to 5 mu g/mL; moderate, 10 mu g/mL; high, 20 to 50 mu g/mL). Attachment and spreading of keratinocytes on fibronectin-coated surfaces in the presence of HNP-1 were evaluated under microscope. Attachment of Fusobacterium nucleatum ATCC25586 and Prevotella intermedia ATCC25611 on keratinocytes preincubated with HNP-1 were determined with a standard antibiotic test. Results: HNP-1 through HNP-3 localized in the junctional epithelium of clinically healthy gingiva and in the pocket epithelium of gingiva with periodontitis. When keratinocyte monolayers were incubated with neutrophil extracts, HNP-1 through HNP-3 were bound to the periphery of the growing cell colonies. In low HNP-1 concentrations, the keratinocyte proliferation enhanced. Moderate and high concentrations of HNP-1 increased the cellular death significantly. HNP-1 increased the attachment and spreading of keratinocytes on fibronectin-coated surfaces and bacterial attachment to keratinocytes in a concentration-dependent manner. Conclusion: HNP-1 plays a role in the integrity of keratinocytes by stimulating their proliferation, attachment, and spreading, whereas higher doses increase the bacterial attachment and keratinocyte death. Salivary IgA and IgG Antibody Responses against Periodontitis-Associated Bacteria in Crohn's Disease

Summary: This study evaluated the levels of IgA and IgG antibodies against Porphyromonas gingivalis, Tannerella forsythia, Aggregatibacter actinomycetemcomitans, and Prevotella intermedia in the saliva of Crohn's disease patients. Higher levels of salivary IgG2 and IgG3 antibodies, as well as IgG antibodies against P. gingivalis, A. actinomycetemcomitans, and P. intermedia, were detected in Crohn's disease patients. The carriage of P. gingivalis in the saliva was lower in Crohn's disease patients.

Serum, saliva, and gingival tissue human β-defensin levels in relation to retinoic acid use

Summary: The study analyzed the effect of systemic retinoic acid use on the levels of human beta-defensin (hBD-1, hBD-2, and hBD-3) in serum, saliva, and gingival tissue. The results showed reduced levels of hBD-2 in saliva of retinoic acid users, but no difference in serum or tissue levels of hBD-1, hBD-2, or hBD-3.

Periodontal microbiology and microbial etiology of periodontal diseases: Historical concepts and contemporary perspectives

Summary: This narrative review provides an overview of the collective knowledge on periodontal microbiology, with a focus on the European contribution in the global field. It discusses the etiological concepts of periodontal disease, particularly the ecological plaque hypothesis and its interpretation. The review also explores anaerobic microbiology, the discovery of periodontal pathogens and their virulence factors, as well as biofilms. Additionally, it highlights the evolution of molecular methods and high-throughput platforms in understanding the periodontal microbiome. Finally, the review examines the role of different microbial species in periodontal diagnosis, the use of microbial and host biomarkers, and the application of antimicrobials in treatment.

Oral Cavity Beta-Defensin Levels Are Regulated Differently during Radiotherapy in Head and Neck Cancer Patients

Summary: This study aimed to investigate the levels of hBD 1-3 in the oral cavity of head and neck cancer patients and its relation to radiotherapy treatment. The results showed that hBD-1 levels increased during radiotherapy but returned to pretreatment levels after the end of radiotherapy. No significant change was detected for hBD-2 or hBD-3 levels during or after radiotherapy.

Salivary Human β-Defensin 1-3 and Human α-Defensin-1 Levels in Relation to the Extent of Periodontal Disease and Tooth Loss in the Elderly

Summary: The aim of this study was to examine the levels of human beta-defensin 1-3 and human neutrophil peptide 1 (HNP-1) in the saliva of elderly individuals, in order to establish the extent of periodontal disease and tooth loss. The results showed that salivary HNP-1 levels were significantly higher in individuals with periodontal disease. The association between salivary HNP-1 levels and the number of pocket teeth remained significant after adjusting for various factors.

Radiotherapy Increases aMMP-8-Levels and Neutrophil/Lymphocyte Ratio Rapidly in Head and Neck Cancer Patients: A Pilot Study

Summary: This study investigated the levels of aMMP-8, molecular forms of MMP-8, blood neutrophil counts and neutrophil/lymphocyte ratios before and after radiotherapy for HNC. The results showed significant increases in aMMP-8 levels and neutrophil/lymphocyte ratios after 3 weeks of radiotherapy. Western immunoblot analysis confirmed elevated levels of activated and fragmented MMP-8 after HNC radiotherapy. These findings suggest an increased risk for periodontitis due to local and systemic inflammation caused by HNC radiotherapy.

Summary: The study aimed to localize MCPIP-1 and MALT-1 in gingival tissues and profile their protein expression levels in relation to inflammation, P. gingivalis colonization, and IL-8 levels. The results showed that MCPIP-1 was present in the epithelium and connective tissue, while MALT-1 was observed in all layers of the gingival epithelium and around inflammatory cells. There was no difference in MCPIP-1 and MALT-1 levels based on gingival inflammation severity, but MALT-1 levels were elevated with an increase in P. gingivalis levels and showed an association with IL-8 levels.

The results showed that salivary concentrations of certain chemokines were elevated and others were decreased in the T1DM group, and the proteolytic activity of MAPKK cleavage was also increased.
