Summary: Both bone marrow mononuclear cells (BMMNCs) and bone marrow stromal cells (BMSCs) have comparable effects on cell repopulation and interfacial allograft-bone healing after allograft anterior cruciate ligament reconstruction (ACLR). Intra-articular delivery of BM cells has limited benefits on graft cell repopulation and may cause higher inflammation in the early stage, with fibrocartilage formation in the tendon-bone interface.

Summary: Osteosarcoma, a bone cancer typically found in adolescents, is closely related to the conversion of bone marrow during adolescence. This study compared the differentiation potential of bone marrow cells from different regions with osteosarcoma cell lines. The results showed distinct differences in the differentiation abilities of hematopoietic and adipocyte-rich bone marrow, which correlated with specific characteristics of the osteosarcoma cell lines.

Summary: In mammals, the interaction between bone marrow stroma and hematopoietic progenitors contributes to bone-BM homeostasis, but the mechanisms and interactions between the development of skeletal and hematopoietic systems are largely unknown. This study reveals that the intracellular O-GlcNAc modification plays a role in dictating the fate and function of early BM stromal cells (BMSCs). O-GlcNAcylation promotes osteogenic differentiation of BMSCs and supports lymphopoiesis, while inhibiting adipogenesis and myelopoiesis. Impaired bone formation and defective hematopoiesis were observed when O-GlcNAc transferase (OGT) was ablated in BMSCs.

Summary: Senescence and altered differentiation potential of bone marrow stromal cells (BMSCs) lead to age-related bone loss. The splicing factor Y-box binding protein 1 (YBX1) regulates BMSC senescence and differentiation shift during aging by controlling RNA splicing. Deletion of Ybx1 in BMSCs accelerates bone loss in mice, while its overexpression stimulates bone formation. Sciadopitysin, a small compound, attenuates the degradation of YBX1 and prevents bone loss in old mice.

AML-mediated activation of Notch signaling is involved in the ineffective osteogenic commitment of BMSCs, resulting in the formation of a pre-osteoblast-rich niche supportive of leukemia development in the bone marrow.

Summary: This study establishes a sustainably releasing system of exosomes-fibrin combinate loaded on tantalum-coating titanium implants. The system has potential effects on osseointegration between tantalum coating titanium implants and the surrounding bone tissue. The study demonstrates that the exosome sustained-release system with fibrin loading on the tantalum coating successfully promotes adhesion, proliferation, and osteogenic differentiation of BMSCs, and significantly enhances the osseointegrative capacity of the implants in vivo.

Summary: This study investigated the effects of multi-walled carbon nanotubes (MWCNTs) on different types of cells in the bone microenvironment. The results showed that MWCNTs promoted osteogenesis in osteoblasts and inhibited osteogenic differentiation in bone marrow stromal cells (BMSCs). In addition, MWCNTs increased bone formation and angiogenesis in the osteolysis model.
