Aims: Micro-ribonucleic acid (miRNA) are noncoding RNA molecules of 21 to 24 nt that regulate the expression of target genes in a post-transcriptional manner. Evidence indicates that miRNA play essential roles in embryogenesis, cell differentiation and pathogenesis of human diseases. This study describes a comparison between the microRNA pro. le of human-papillomavirus-negative cervical squamous cell carcinoma patients and controls, in order to develop further understanding of the pathogenesis of cervical squamous cell carcinomas. Methods: MiRNA were isolated from tumor tissues of five human-papillomavirus-negative cervical squamous cell carcinoma patients and five healthy controls in order to perform miRNA microarray chip analysis. The chip results were then confirmed by northern blot analysis. Results: A total of 27 miRNA differentially expressed between the squamous cell carcinoma patients and the healthy controls were identified. Conclusion: This work indicates that these miRNA may be potential diagnosis biomarkers and probable factors involved in the pathogenesis of cervical squamous cell carcinomas.

Single-Cell RNA and ATAC Sequencing Reveal Hemodialysis-Related Immune Dysregulation of Circulating Immune Cell Subpopulations

Hongwei Wu, Jingjing Dong, Haiyan Yu, Kang Wang, Weier Dai, Xinzhou Zhang, Nan Hu, Lianghong Yin, Donge Tang, Fanna Liu, Yong Dai

Summary: This study integrated single-cell RNA sequencing (scRNA-seq) and single-cell assay for transposase-accessible chromatin sequencing (scATAC-seq) to investigate the transcriptional program and chromosomal accessibility of peripheral mononuclear cells (PBMCs) in maintenance hemodialysis patients. The study found that hemodialysis suppressed the expression of immune-related genes in CD4+ T cells and monocytes, inhibited downstream pathways of T cell receptor signaling, and altered cellular communication patterns between PBMC subgroups. Additionally, hemodialysis affected the chromatin accessibility profile and expression of transcription factors. This study provides a valuable framework for studying hemodialysis-related immune dysregulation and identifies potential therapeutic targets.

Summary: This study investigated the biological function of circSHKBP1 in NSCLC and found that it enhanced proliferation, migration, invasion, and stemness of NSCLC cells. The study also identified the miR-1294/PKM2 axis as the mechanism by which circSHKBP1 regulates NSCLC progression. These findings suggest that circSHKBP1 may serve as a potential prognostic biomarker and therapeutic target for NSCLC.

New insights for regulatory T cell in lupus nephritis

Yixi Li, Donge Tang, Lianghong Yin, Yong Dai

Summary: Lupus nephritis (LN) is a complex autoimmune disease, and recent research has shown the potential of regulatory T (Treg) cell therapy in treating autoimmune diseases. However, there is limited understanding of Treg cell therapy for LN patients. This review aims to summarize new insights into Treg-targeting techniques in LN patients.

Analyzing the gene regulatory network in hepatitis B patients by single-cell ATAC sequencing

Huixuan Xu, Haiyan Yu, Fengping Zheng, Cantong Zhang, Wanxia Cai, Xinzhou Zhang, Donge Tang, Yong Dai

Summary: This study provides a new perspective for understanding the pathophysiology of chronic hepatitis B (CHB) by analyzing the gene regulatory network in CHB patients using single-cell ATAC sequencing. The specific cell types associated with CHB were found to be located in B-0 and T-3. The regulatory network of the hepatitis B signal pathway composed of genes linked to the differentially expressed peaks of these two CHB disease-specific cell types was drawn.

Integrated single-cell analyses decode the developmental landscape of the human fetal spine

Haiyan Yu, Donge Tang, Hongwei Wu, Chunhong Li, Yongping Lu, Fang He, Xiaogang Zhang, Yane Yang, Wei Shi, Wenlong Hu, Zhipeng Zeng, Weier Dai, Minglin Ou, Yong Dai

Summary: This study used integrated single-cell RNA sequencing and single-cell transposase-accessible chromatin sequencing analyses to investigate cellular heterogeneity and transcriptional regulatory networks during human spine development. The findings revealed that fibroblasts with stem cell characteristics can differentiate into chondrocytes and neurons may originate from neuroendocrine cells. The interaction between macrophages and neurons involving NRP2_SEMA3C and CD74_APP is crucial for spinal cord development.

Summary: According to the research, the reduced expression of GSTM2 in colorectal cancer is associated with lower overall survival rate of patients. The study uncovered potential regulatory mechanisms of GSTM2 in colorectal cancer, including deletion on the genome and up-regulation of other genes. Moreover, GSTM2 might be involved in immune-related pathways and regulate the development of tumors by modulating the immune microenvironment.

Summary: By analyzing the microbiota and serum metabolites in urine samples from patients with diabetic kidney disease (DKD), it was found that DKD patients had a different urinary microbiota compared to the healthy controls, with the most abundant microbiota being Proteobacteria and Acidobacteria. Short-chain fatty acids and protein-bound uremic toxins were shown to be specific to DKD. Furthermore, the study revealed that the metabolism of arginine and proline may play a major role in the regulation of DKD.
