Comparison of 99mTc- and 18F-ubiquicidin autoradiography to anti-Staphylococcus aureus immunofluorescence in rat muscle abscesses

Tc-99m-ubiquicidin (UBI) 29-41 is under clinical evaluation for discrimination between bacterial infection and unspecific inflammation. We compared the distribution of Tc-99m-UBI 29-41, the potential PET tracers F-18-UBI 29-41 and F-18-UBI 28-41, and H-3-deoxyglucose (DG) in rat muscle abscesses to that of anti-Staphylococcus aureus immunofluorescent imaging. Methods: Calf abscesses were induced in 15 CDF-Fischer rats after inoculation of Staphylococcus aureus. One to 6 d later, either F-18-UBI 29-41 and H-3-DG (n = 5) or F-18-UBI 28-41 and H-3-DG (n = 6) or Tc-99m-UBI 29-41 and H-3-DG (n = 4) were injected simultaneously. Dual-tracer autoradiography of the abscess area was compared with the distribution of bacteria and macrophages. Results: The UBI derivates exhibited increased uptake in the abscess area that partly matched H-3-DG uptake and macrophage infiltration but showed no congruity with areas that were highly positive for bacteria. Conclusion: A specific binding of UBI derivatives to Staphylococcus aureus in vivo could not be confirmed in this study.

Lesion-Function Analysis from Multimodal Imaging and Normative Brain Atlases for Prediction of Cognitive Deficits in Glioma Patients

Summary: This study used structural MRI and amino acid PET in conjunction with brain atlases and elastic registration techniques to investigate the impact of treatment-related brain damage on cognitive functioning in well-functioning WHO Grade III/IV glioma patients. The results showed that T2/FLAIR hyperintensities caused by radiation therapy significantly impaired cognitive performance, especially affecting the left hemisphere functional nodes and fiber tracts, which were highly correlated with verbal episodic memory dysfunction.

A multimodal approach that combines PET and MRI shows promising improvement in the diagnosis of cerebral gliomas.

A Linearized Fit Model for Robust Shape Parameterization of FET-PET TACs

Summary: A linearized model for dynamic PET scans of F-18-FET TACs was derived and validated in this study, showing high agreement with measured TACs and small parameter uncertainties. The model demonstrated good diagnostic performance in discriminating between IDH wildtype and IDH mutant gliomas in adult patients. Voxel-wise fitting within a short computation time and comparable classification accuracy to linear fits based on average TAC values were achievable.
