Comprehensive analysis of neurobehavior associated with histomorphological alterations in a chronic constrictive nerve injury model through use of the Cat Walk XT system

Object. Neuropathic pain is debilitating, and when chronic, it significantly affects the patient physically, psychologically, and socially. The neurobehavior of animals used as a model for chronic constriction injury seems analogous to the neurobehavior of humans with neuropathic pain. However, no data depicting the severity of histomorphological alterations of the nervous system associated with graded changes in neurobehavior are available. To determine the severity of histomorphological alteration related to neurobehavior, the authors created a model of chronic constrictive injury of varying intensity in rats and used the CatWalk XT system to evaluate neurobehavior. Methods. A total of 60 Sprague-Dawley rats, weighing 250-300 g each, were randomly assigned to 1 of 5 groups that would receive sham surgery or 1, 2, 3, or 4 ligatures of 3-0 chromic gut loosely ligated around the left sciatic nerve. Neurobehavior was assessed by CatWalk XT, thermal hyperalgesia, and mechanic allodynia before injury and periodically after injury. The nerve tissue from skin to dorsal spinal cord was obtained for histomorphological analysis 1 week after injury, and brain evoked potentials were analyzed 4 weeks after injury. Results. Significant differences in expression of nerve growth factor existed in skin, and the differences were associated with the intensity of nerve injury. After injury, expression of cluster of differentiation 68 and tumor necrosis factor a was increased, and expression of S100 protein in the middle of the injured nerve was decreased. Increased expression of synaptophysin in the dorsal root ganglion and dorsal spinal cord correlated with the intensity of injury. The amplitude of sensory evoked potential increased with greater severity of nerve damage. Mechanical allodynia and thermal hyperalgesia did not differ significantly among treatment groups at various time points. CatWalk XT gait analysis indicated significant differences for print areas, maximum contact maximum intensity, stand phase, swing phase, single stance, and regular index, with sham and/or intragroup comparisons. Conclusions. Histomorphological and electrophysiological alterations were associated with severity of nerve damage. Subtle neurobehavioral differences were detected by the CatWalk XT system but not by mechanical allodynia or thermal hyperalgesia. Thus, the CatWalk XT system should be a useful tool for monitoring changes in neuropathic pain, especially subtle alterations.

Aggravation of pulmonary fibrosis after knocking down the aryl hydrocarbon receptor in the insulin-like growth factor 1 receptor pathway

Summary: This study investigated the cascade pathway involving IGF1R and AhR in the development of idiopathic pulmonary fibrosis. The results showed increased IGF1R levels and decreased AhR expression in idiopathic pulmonary fibrosis patients and animal models. Knockout of AhR aggravated lung fibrosis, while the use of IGF1R inhibitor and AhR agonist attenuated such effects. TGF-beta1 and BLM suppressed AhR expression through endoplasmic reticulum stress and activated IGF1R. This study provides a potential target for the treatment of lung injury and fibrosis.

Targeting histone deacetylase-3 blocked epithelial-mesenchymal plasticity and metastatic dissemination in gastric cancer

Summary: This study found that Honokiol can prevent the metastatic dissemination of gastric cancer cells by inhibiting HDAC3 activity/expression. The results suggest that Honokiol inhibits HDAC3 activity by suppressing the activation of the NF-Bp65/CEBPβ signaling pathway, thereby inhibiting the migration and invasion of cancer cells. In addition, Honokiol increases the expression of ER stress markers, inhibits the expression of EMT-associated epithelial markers, and decreases Wnt/β-catenin activity, thereby blocking the metastasis of gastric cancer.

ANGPTL1 attenuates cancer migration, invasion, and stemness through regulating FOXO3a-mediated SOX2 expression in colorectal cancer

Summary: This study identified that ANGPTL1 is down-regulated in colorectal cancer (CRC) and is inversely correlated with metastasis and poor clinical outcomes. ANGPTL1 suppresses migration/invasion abilities, the expression of cancer stem cell markers, and sphere formation in CRC cells by enhancing FOXO3a expression. Overexpression of ANGPTL1 reduces liver metastasis, tumor growth, and tumorigenicity in tumor-bearing mice.

Gamma Knife Radiosurgery for Indirect Dural Carotid-Cavernous Fistula: Long-Term Ophthalmological Outcome

Summary: The study investigates the regulatory mechanism of the AHR/TET2/NT5E pathway during SLE progression. The expression levels of AHR, TET2, and NT5E were examined in Tregs of SLE patients. The study found that AHR and TET2 expression levels were correlated with the immunosuppressive functions of Tregs. In addition, KYN promoted the binding of AHR to the TET2 promoter, leading to downregulation of NT5E expression in Tregs of SLE patients. Furthermore, the study showed that the Treg immunosuppressive activity was enhanced in the KYN-treated group through the TET2 and A2AR-adenosine pathways.

Modulation of Aryl Hydrocarbon Receptor Expression Alleviated Neuropathic Pain in a Chronic Constriction Nerve Injury Animal Model

Summary: Neuropathic pain is reduced by AhR activation and aggravated by AhR deletion, indicating the neuroprotective effects of AhR. AhR expression is increased after nerve injury, and AhR agonist omeprazole attenuates nerve damage and inflammation. AhR agonists may have potential in neuropathic pain treatment.

Melatonin protects retinal integrity through mediated immune homeostasis in the sodium iodate-induced mouse model of age-related macular degeneration

Summary: Age-related macular degeneration is a major cause of visual deficiency in older adults worldwide. Melatonin may reduce retinal deterioration by mediating regulatory T cells in the retina. The study found that melatonin effectively restored retinopathy and maintained retinal structural integrity. It also promoted tissue repair by increasing the infiltration of regulatory T cells and upregulating TET2 and NT5E expression.

Using the deformity index of vital structures to predict outcome of patients with large vestibular schwannomas after Gamma Knife radiosurgery

Summary: This study aimed to use automated volumetric analysis software to quantify the degree of brain stem deformity and predict the long-term outcomes of patients with large vestibular schwannomas (VS) following Gamma Knife radiosurgery (GKRS). The study found that patients with smaller tumor volumes, lower vital structure deformity indices, and a longer distance of the tumor to the central line were more likely to have favorable clinical outcomes after GKRS. The brainstem deformity ratio is likely a useful index to assess the clinical and tumor regression outcomes.

Thrombin-Induced Microglia Activation Modulated through Aryl Hydrocarbon Receptors
