Summary: This study found that alpha-2A adrenoceptors (alpha 2-AR) in the nervous system of streptozotocin diabetic rats are upregulated, and the upregulation is associated with increased pain sensitivity. Chronic administration of the selective alpha 2-AR agonist guanfacine can downregulate the upregulation of alpha 2-AR and relieve hyperalgesia and allodynia in diabetic animals. These findings suggest that guanfacine may have potential as a potent analgesic for diabetic neuropathic pain.

The system is able to evaluate footfall and gait changes, calculate various parameters, and suggest new therapies based on standardized methods for regeneration of peripheral nerve and spinal cord lesions.

Summary: The study investigated the anti-osteoarthritic effects of the herbo-mineral formulation Peedanil Gold (PN-G) in a rat model of osteoarthritis induced by monosodium-iodoacetate injection. PN-G treatment improved clinical scores, reduced hyperalgesia and allodynia, ameliorated joint inflammation, and promoted cartilage regeneration. It also showed dose-dependent anti-inflammatory effects by reducing the levels of IL-6 and IL-1 beta in inflamed human macrophagic THP-1 cells. The presence of various analgesic and anti-inflammatory phytocompounds in PN-G was confirmed by UHPLC analysis.

Summary: This study investigated the cascade pathway involving IGF1R and AhR in the development of idiopathic pulmonary fibrosis. The results showed increased IGF1R levels and decreased AhR expression in idiopathic pulmonary fibrosis patients and animal models. Knockout of AhR aggravated lung fibrosis, while the use of IGF1R inhibitor and AhR agonist attenuated such effects. TGF-beta1 and BLM suppressed AhR expression through endoplasmic reticulum stress and activated IGF1R. This study provides a potential target for the treatment of lung injury and fibrosis.

Summary: This study found that Honokiol can prevent the metastatic dissemination of gastric cancer cells by inhibiting HDAC3 activity/expression. The results suggest that Honokiol inhibits HDAC3 activity by suppressing the activation of the NF-Bp65/CEBPβ signaling pathway, thereby inhibiting the migration and invasion of cancer cells. In addition, Honokiol increases the expression of ER stress markers, inhibits the expression of EMT-associated epithelial markers, and decreases Wnt/β-catenin activity, thereby blocking the metastasis of gastric cancer.

Summary: The study investigates the regulatory mechanism of the AHR/TET2/NT5E pathway during SLE progression. The expression levels of AHR, TET2, and NT5E were examined in Tregs of SLE patients. The study found that AHR and TET2 expression levels were correlated with the immunosuppressive functions of Tregs. In addition, KYN promoted the binding of AHR to the TET2 promoter, leading to downregulation of NT5E expression in Tregs of SLE patients. Furthermore, the study showed that the Treg immunosuppressive activity was enhanced in the KYN-treated group through the TET2 and A2AR-adenosine pathways.

Summary: This study aimed to use automated volumetric analysis software to quantify the degree of brain stem deformity and predict the long-term outcomes of patients with large vestibular schwannomas (VS) following Gamma Knife radiosurgery (GKRS). The study found that patients with smaller tumor volumes, lower vital structure deformity indices, and a longer distance of the tumor to the central line were more likely to have favorable clinical outcomes after GKRS. The brainstem deformity ratio is likely a useful index to assess the clinical and tumor regression outcomes.
