Optimal Preclinical Conditions for Using Adult Human Multipotent Neural Cells in the Treatment of Spinal Cord Injury

Summary: Stem cell-based therapeutics, particularly adult human multipotent neural cells (ahMNCs) from patients with hemorrhagic stroke, show potential in promoting significant locomotor recovery in spinal cord injury (SCI) through modulating glial scar formation, neuroprotection, and/or angiogenesis. The optimal transplantation dose of ahMNCs identified in this study could be beneficial in determining the appropriate injection dose for SCI patients in the future.

Lasmiditan efficacy in migraine attacks with mild vs. moderate or severe pain

Summary: The study found that treating migraine attacks with LTN at mild pain intensity resulted in numerically greater 2-hour pain freedom and 24-hour sustained pain freedom response rates compared to attacks treated at moderate or severe pain intensity. Attacks treated at mild pain also had a significantly higher proportion of achieving 2-hour pain freedom and most bothersome symptom freedom, as well as 24-hour sustained pain freedom.

Adult Human Multipotent Neural Cells Could Be Distinguished from Other Cell Types by Proangiogenic Paracrine Effects via MCP-1 and GRO

Summary: Adult human multipotent neural cells (ahMNCs) derived from adult human temporal lobes exhibit unique characteristics, including distinctive cell marker expression, higher neural differentiation potentials, and increased expression of paracrine factors. These features set ahMNCs apart from other cell types such as pericytes, mesenchymal stem cells (MSCs), and fetal neural stem cells (fNSCs). The distinct properties of ahMNCs make them valuable for potential applications in regenerative medicine research and as comparative references for other cell types.

The digiti quinti sign in hemiplegic migraine: An fMRI study

Summary: This study demonstrated the presence of Digiti quinti sign (DQS) in hemiplegic migraine patients, indicating a permanent subtle motor dysfunction and disrupted cortical activation. Could Machine Learning Better Predict Postoperative C5 Palsy of Cervical Ossification of the Posterior Longitudinal Ligament?

Summary: This study aimed to investigate whether machine learning (ML) can predict postoperative C5 palsy of cervical ossification of the posterior longitudinal ligament (OPLL) patients better than a conventional logistic regression. Four sampling methods were used to balance the data, and six ML algorithms and conventional logistic regression were compared. The results showed that the ML algorithm using adaptive reinforcement learning and downsampling had the highest AUC and outperformed logistic regression. Age, surgical method, and involvement of OPLL at C1-C3 were significantly associated with C5 palsy, and postoperative shoulder pain was closely related to postoperative C5 palsy of OPLL patients.

Long-term Follow-up Results of Reconstructive Laminoplasty With L-shaped Leibinger Mini-plate for Posterior Approach in the Treatment of Intraspinal Tumor Surgery

Summary: This study aimed to determine the long-term clinical and radiologic outcomes using L-shaped mini-plates for laminoplasty. The results showed that laminoplasty using L-shape Leibinger mini-plates had a fusion rate of 89.3% and no displacement of the re-attached laminae was observed. It is considered a safe and feasible option for surgeries for intraspinal pathologies.

Sustained response of galcanezumab in migraine prevention: Patient--level data from a post hoc analysis in patients with episodic or chronic migraine

Prevalence of chronic pain in Brazil: A systematic review and meta-analysis

The study found that chronic pain was highly prevalent in the general adult population, with a range of 23.02% to 41.4%, and in the elderly population, with a range of 29.3% to 76.2%. Chronic pain was associated with various factors such as gender, age, education level, lifestyle choices, and mental health.

JAK2/STAT3 pathway mediates neuroprotective and pro-angiogenic treatment effects of adult human neural stem cells in middle cerebral artery occlusion stroke animal models

Summary: This study demonstrated the neuroprotective and pro-angiogenic effects of adult human neural stem cells (ahNSCs) in the treatment of ischemic stroke. Tissue analysis showed that ahNSCs reduced brain tissue atrophy and functional loss, and preserved neurons by decreasing reactive astrogliosis and microglial activation. Furthermore, these effects were mediated by paracrine factors and reversed when the JAK2/STAT3 signaling pathway was inhibited. This preclinical study provides important insights for the development of stem cell therapeutics for ischemic stroke.

Summary: The study found that long-term in vitro expansion of adult human multipotent neural cells (ahMNCs) can induce copy number variations and genetic mutations, but the number did not increase with culture progression and many of them disappeared. Although most of the observed genetic mutations were not shared by all ahMNCs, non-synonymous missense mutations at the MUC4 gene were found in three out of five long-term cultured ahMNC lines. The genetic instability did not confer in vivo tumorigenic potential to ahMNCs.
