Summary: Immunofluorescence staining is widely used in the subcellular localization study of proteins. A tissue-chopping-based method for immunofluorescence staining of chloroplast proteins is improved by direct observation of stained tissues using a confocal microscope without tissue lysis and by maintaining a low temperature throughout the process. This method has been successfully applied to the staining of FtsZ1 in wildtype and mutant Arabidopsis, as well as in other plants, leading to further improvement of the tissue-chopping-based immunofluorescence staining method.

Summary: Fluorescence imaging of the whole brain is crucial for comprehensive studies of brain structure and function. However, obtaining high quality immunofluorescence staining on cleared samples has been challenging. In this study, we developed TSA-PACT, a method combining tyramide signal amplification (TSA) and PACT, to improve sensitivity, specificity, and stability of immunofluorescence signals in the whole brain of zebrafish.

Summary: Many funding agencies have emphasized the importance of considering sex as a biological variable in experimental design to improve the reproducibility and translational relevance of preclinical research. Omitting the female sex from experimental designs in neuroscience and pharmacology can result in biased or limited understanding of disease mechanisms. This article provides methodological considerations for incorporating sex as a biological variable in in vitro and in vivo experiments, including the influence of age and hormone levels, and proposes strategies to enhance methodological rigor and translational relevance in preclinical research.
