Summary: The expression of Receptor activator of NF-kappa B (RANK) is increased in podocytes of patients with diabetic nephropathy. Mouse models of podocyte-specific RANK depletion and overexpression showed that RANK depletion alleviated the development of kidney diseases, while RANK overexpression aggravated the condition. The findings suggest that RANK mediates the development of diabetic nephropathy by promoting glomerular oxidative stress and proinflammatory cytokine production.

Summary: Gastric hypersensitivity is a major feature of functional dyspepsia (FD) and many patients with FD exhibit gastroduodenal microinflammation. This study used a maternal separation-induced FD model to demonstrate the functional link between gastroduodenal microinflammation and gastric hypersensitivity. The findings suggest that anti-inflammatory therapy may be effective in treating FD patients with gastroduodenal microinflammation.

Summary: The aim of this study was to investigate the mechanism and interaction of miR-181a, TLR4, and NF-kappa B in gastric hypersensitivity in diabetic rats. The expression of TLR4 was up-regulated in the DRGs of diabetic rats, while miR-181a and miR-7a were down-regulated. MiR-181a directly regulated TLR4 expression and reduced the hyper-excitability of gastric-specific neurons, thus alleviating gastric hypersensitivity.

Summary: This study investigates the potential role of ANP32E in the development of gastric cancer. The results demonstrate that ANP32E is overexpressed in gastric cancer tissues and its overexpression promotes malignancy while its knockdown inhibits proliferation and induces apoptosis of gastric cancer cells. ANP32E is also found to be positively associated with NUF2 expression and shown to induce NUF2 expression, promoting tumor cell proliferation and inhibiting apoptosis in gastric cancer cell lines.

Summary: This study aimed to investigate the protective role of mimetic peptide Ac2-26 of annexin A1 in regulating p38MAPK/NF-Kappa B in a diabetic mouse model. The results showed that treatment with Ac2-26 significantly improved fasting blood glucose, blood lipid levels, and renal function, while reducing body weight in the Ac2-26 group compared to the model group. The levels of inflammatory factors in renal tissue were also significantly decreased in the Ac2-26 group. Moreover, the mRNA and protein expressions of p38MAPK, NF-Kappa BP6, and ANXA1 were significantly reduced in the Ac2-26 group.

Summary: Inflammatory and immune pathways are crucial in the pathophysiology of non-alcoholic fatty liver disease (NAFLD), and Sitagliptin's role in altering these pathways to improve NAFLD was investigated in this study. The results demonstrated that Sitagliptin can ameliorate NAFLD by decreasing HMGB1-mediated TLR4/NF-kappa B signaling to suppress inflammation and reduce insulin resistance, suggesting its potential as a therapeutic intervention for NAFLD.
