Summary: Mucuna pruriens seed extract demonstrated significant anti-inflammatory effects in LPS-stimulated BV2 microglial cells by inhibiting the release of inflammatory mediators such as NO, IL-1 beta, IL-6, and TNF-alpha, and by suppressing the NF-kappa B signaling pathway. These findings suggest the potential use of M. pruriens seeds in traditional and alternative medicine for treating PD and other inflammation-associated diseases.

Summary: High glucose favors NO production, but pro-inflammatory cytokine production is highest in the presence of carbon sources that drive OXPHOS. Glutamine is a very potent inducer of IL6 production among carbon sources that drive OXPHOS, and this effect is dampened in the presence of glucose. These findings may offer new prospects for the therapeutic manipulation of neuroinflammation in the context of diabetes and AD.

Summary: Abnormal infiltration and activation of neutrophils contribute to lupus nephritis (LN) development. Myeloid-related proteins (MRPs), MRP-8, and -14, known as DAMPs, are secreted by activated neutrophils in systemic lupus erythematosus (SLE). Mesenchymal stem cells (MSCs) have the ability to regulate immune cells and treat LN, but their effect on neutrophils and MRP-8/14 expression in LN is unclear. This study showed that MSC transplantation reduced neutrophil infiltration and MRP-8/14 expression in the kidneys of MRL/lpr mice. Furthermore, it was found that TSG-6 secreted by MSCs played a crucial role in inhibiting MRP-8/14 expression in neutrophils and neutrophil migration. Immunosuppressants had no significant effect on MRP-8/14 expression in neutrophils. Thus, MSCs can inhibit MRP-8/14 expression and neutrophil migration in LN through the secretion of TSG-6.

Summary: Microglia-mediated neuroinflammation is crucial in the pathophysiology of neurodegenerative diseases. Lipopolysaccharide (LPS) can activate microglia and induce inflammation. Matrix metalloproteinases (MMPs) and CD147 are key factors involved in microglial activation and inflammation. This study found that CD147 expression was induced in LPS-induced microglial activation, and inhibiting CD147 expression could suppress microglial activation. Furthermore, MMPs and autophagy were involved in the inflammatory activation of microglia, and inhibiting MMPs or autophagy could attenuate this activation.
