Summary: Activated Cdc42-associated kinase 1 (ACK1/TNK2) is a nonreceptor tyrosine kinase with a unique structure that can act as a transmembrane effector of receptor tyrosine kinases and play a role in epigenetic regulation. ACK1 has been shown to be a carcinogenic factor, and blocking it can inhibit cancer cell survival, proliferation, migration, and radiation resistance. Despite efforts to develop ACK1 inhibitors, no specific small molecule inhibitors have entered clinical trials yet.

Summary: Phosphatases and kinases play crucial roles in maintaining the balance between dephosphorylated and phosphorylated proteins for cellular functions. Imbalance or dysfunction in their activities can lead to unfavorable cellular effects and the development of diseases. Protein tyrosine phosphatases (PTPs) are attractive therapeutic targets due to their involvement in cell signaling and diseases, but they have been challenging in drug development. This article focuses on the progress in small-molecule inhibition of the mitogen-activated protein kinase (MAPK) phosphatases (MKPs), discussing successful strategies and inhibitor discovery tools, as well as the potential future of MKP inhibition.

Summary: It was found that extracellular acidosis (EA) significantly alters the gene expression of intestinal epithelial cells (IEC) as an adaptive mechanism against inflammation through a series of molecular mechanisms. In murine and human ileitis inflammation-associated acidosis, a correlation was observed between the expression of acidosis-related genes and bacterial reporter gene intensity.
