Strong therapeutic potential of γ-secretase inhibitor MRK003 for CD44-high and CD133-low glioblastoma initiating cells

The Notch signal regulates both cell viability and apoptosis, and maintains stemness of various cancers including glioblastoma (GBM). Although Notch signal inhibition may be an effective strategy in treating GBM initiating cells (GICs), its applicability to the different subtypes of GBM remains unclear. Here, we analyzed the effectiveness of MRK003, a preclinical gamma-secretase inhibitor, on GICs. Nine patient-derived GICs were treated by MRK003, and its efficacy on cell viability, apoptosis, sphere forming ability and Akt expression level which might be related to Notch downstream and be greatly important signals in GBM was evaluated. MRK003 suppressed viability and sphere-formation ability, and induced apoptosis in all GICs in varying doses of MRK003. Based on their sensitivities to MRK003, the nine GICs were divided into relatively sensitive'' and relatively resistant'' GICs. Sensitivity to MRK003 was associated with its inhibitory effect on Akt pathway. Transgenic expression of the myristoylated Akt vector in relatively sensitive GICs partially rescued the effect of MRK003, suggesting that the effect of MRK003 was, at least in part, mediated through inhibition of the Akt pathway. These GICs were differentiated by the expression of CD44 and CD133 with flow cytometric analysis. The relatively sensitive GICs are CD44-high and CD133-low. The IC50 of MRK003 in a set of GICs exhibited a negative correlation with CD44 and positive correlation with CD133. Collectively, MRK003 is partially mediated by the Akt pathway and has strong therapeutic potential for CD44-high and CD133-low GICs.

Minimum wall shear stress points and their underlying intra-aneurysmal flow structures of unruptured cerebral aneurysms on 4D flow MRI

Summary: This study investigated the relationship between the locations of minimum wall shear stress (Min-WSS) points and their underlying intra-aneurysmal flow structure patterns in unruptured cerebral aneurysms using four-dimensional flow magnetic resonance imaging (MRI). The majority of Min-WSS points were found to be located around a vortex core or center, and different locations had different Min-WSS values. Further research can help identify cerebral aneurysms with a higher risk of wall instability.

Tumorigenic activation around HPV integrated sites in head and neck squamous cell carcinoma

Summary: Human papillomavirus (HPV) plays a causal role in the development of head and neck squamous cell carcinoma (HNSCC) by integrating into the host genome and causing genomic alterations. This study analyzed the epigenome, transcriptome, and interactome of HPV-positive HNSCCs to understand the role of epigenetic dysregulation in tumorigenesis. The results showed that genes neighboring HPV integration sites were upregulated and correlated with oncogenic phenotypes in HPV-positive HNSCCs. The epigenetic activation of HPV-interacting regions (HPVIRs) contributed to the genesis of HPV-positive HNSCC.

Patterns of failure in glioblastoma multiforme following Standard (60 Gy) or Short course (40 Gy) radiation and concurrent temozolomide

The effect of damage to the white matter network and premorbid intellectual ability on postoperative verbal short-term memory and functional outcome in patients with brain lesions

Summary: This study investigated whether a higher premorbid IQ has a beneficial impact on postoperative verbal short-term memory and functional outcomes in patients with brain tumors. The results showed that low white matter tract damage and low premorbid IQ both had negative effects on postoperative functional outcomes. However, when the brain lesions were not located in a crucial pathway, a higher premorbid IQ was associated with better postoperative verbal short-term memory and functional outcomes.

Evaluation of a scoring system to assess proficiency in cerebral angiography for neuroendovascular surgery education

Liquid biomarkers in glioma

Summary: An ideal biomarker for glioma must meet certain parameters, but the nature of the disease makes it challenging. Liquid biomarkers for brain disease offer advantages over traditional histological examination. However, the heterogeneous nature of glioma and the low incidence of brain tumors hinder progress in discovering useful biomarkers. New advancements in specimen types and comprehensive omics analysis have identified some candidate liquid biomarkers. Further research and multicenter studies are needed to establish new treatment strategies.

Therapeutic potential of pentamidine for glioma-initiating cells and glioma cells through multimodal antitumor effects

Pentamidine inhibited proliferation and stemness of glioma-initiating cells, and showed anticancer effects on differentiated glioma-initiating cells and glioblastoma cell lines. The in vivo study confirmed the anticancer effects of pentamidine on glioma cells.

Factors related to high bifurcation level of common femoral artery

Summary: This study investigated the level of bifurcation in the common femoral artery (CFA) in 100 patients and found that approximately one-fourth of the patients had a high level of CFA bifurcation, which was associated with old age and a history of diabetes. For patients in need of urgent treatment, femoral artery puncture can be performed at a higher level below the inguinal ligament if they are elderly or have a history of diabetes.

Sympathetic Nervous Hyperactivity Impairs Microcirculation Leading to Early Brain Injury After Subarachnoid Hemorrhage

Summary: This study investigated the role of cerebral circulation in the acute phase following SAH using patient data and a mouse SAH model, and evaluated its regulation via the sympathetic nervous system. The results showed that cerebral circulation time was prolonged in the acute phase of SAH, especially in patients with electrocardiographic changes. In the mouse study, sympathetic hyperactivity was found to impair cerebral microcirculation and contribute to brain edema development in the acute phase following SAH.

Optical coherence tomography detects early optic nerve damage before visual field defect in patients with pituitary tumors

Summary: Optical coherence tomography (OCT) is a useful tool for predicting visual recovery after pituitary tumor removal, but its utility in patients with normal visual fields is unclear. This study analyzed OCT features in pituitary tumors without visual field defects and found that macular thickness ganglion cell complex (mGCC) was significantly thinner in patients with chiasmal compression (CC) compared to those without. Abnormal mGCC thickness was more common in the CC group and associated with older age. OCT can detect early optic nerve damage in patients with pituitary tumors, even in the absence of visual field defects.

Cancer among children, adolescents and young adults in the Hokushin region, Japan, between 2010 and 2015

Summary: The study analyzed cancer data of children, adolescents, and young adults in four adjacent prefectures of Japan, revealing differences in cancer incidence rates among different age groups. Carcinomas were found to be the most common type of cancer in the 15-39 age group but not in the pediatric population. Further research is needed to understand the unique needs of adolescents and young adults with cancer.
