Summary: The study revealed the pathways and signals in the bulge that control repigmentation in human vitiligo. Through selective capture and RNA-sequencing of cells from the hair follicle bulge, it was found that signals like KCTD10, CTNNB1, and RHO play a role in repigmentation, laying the foundation for the discovery of molecules that induce robust repigmentation in vitiligo.

Summary: The synergistic role of Foxp1/4 in maintaining the quiescence of HFSCs and the stem cell niche is revealed. The double deficiency of Foxp1/4 leads to premature activation of HFSCs and hair loss, accompanied by the destruction of the stem cell niche.

Summary: The role of fatty acid desaturation catabolized by SCD1 in regulating hair follicle stem cells and hair growth has been discovered. The absence of SCD1 leads to abnormal hair growth, affecting the appearance and quality of hair.

Summary: This study investigates the molecular heterogeneity of hair follicle stem cells (HFSCs) using high-resolution single-cell RNA sequencing and identifies five major populations distinguished by known markers associated with the bulge and outer root sheath (ORS). Lineage trajectory analysis predicts prospective differentiation paths of cellular states within these populations. Gene set enrichment analysis reveals potential distinct functions of different subpopulations within the bulge and ORS, shedding light on communication and nutrient transfer mechanisms within the hair follicle.

Summary: Complete skin reconstruction is a complex process involving regeneration and reconstruction of various components. In this study, 3D printed Cu-BG/CS wound dressings were designed, which exhibited antibacterial properties, vascular growth, and improved wound healing and functional repair. The results demonstrated that Cu-BG/CS@SB dressings can be used for skin wound healing and hair follicle regeneration.

Summary: Developmental TE is a novel paradigm aiming to obtain tissues and organs with correct biomorphology and biofunctionality by mimicking the morphogenetic processes leading to the tissue/organ generation in the embryo. Successful applications have been reported for ectodermal appendages such as teeth, hair follicles and glands, with a focus on accurate selection of cell sources, scaffolds, and cell culture configurations to replicate in vivo morphogenetic cues.

Summary: Severe skin damage often requires the transplantation of artificial skin substitutes, but these repaired parts may not be able to regenerate accessory organs like hair follicles. Research on artificial skin substitutes that can promote hair follicle regeneration is crucial. Through the use of dermal papilla cells cultured on a biomimetic silk fibroin/sodium alginate scaffold, it was found that this scaffold can effectively promote skin repair and induce hair follicle regeneration in animal models.

Summary: This study investigated the effects of a polycaprolactone scaffold and hair follicle stem cells (HFSCs) on the healing of incisional cutaneous wounds in streptozotocin-induced diabetic rats. The results showed that HFSCs and the scaffold promoted angiogenesis, restored diabetes-induced dysfunction, and accelerated wound closure in diabetic rats.

Summary: The aim of this study was to investigate the effects of maternal thyroid dysfunction on the development of the skin and its derivatives in their offspring in the early postnatal period. The main findings were that both forms of maternal hypothyroidism lead to serious damage of the epidermis in pups, characterized by pronounced hyperkeratosis and reduction of the germinal layer, along with a reduced number of hair follicles and delayed morphogenesis.

Summary: Efficient local delivery of mesenchymal stem cells (MSCs) is essential for their use in tissue regeneration. In this study, a biomimetic bilayer silk fibroin/sodium alginate (SF/SA) scaffold was developed to deliver human umbilical cord mesenchymal stem cells (hUC-MSCs) for wound healing. The scaffold demonstrated favorable properties in vitro and the hUC-MSCs exhibited good viability and stemness. In vivo evaluation showed that the SF/SA scaffolds loaded with hUC-MSCs promoted scarless skin healing and hair follicle regeneration by regulating various biological processes and suppressing endoplasmic reticulum stress. This study highlights the potential of the bilayer SF/SA scaffold as an effective delivery platform for hUC-MSCs and its therapeutic application in tissue regeneration.

Summary: The preparation of biodegradable scaffolds loaded with cells and cytokine is a feature of tissue-engineered skin. IPSCs-based tissue-engineered skin treatment for wound repair is worth exploring. CK19(+)/Integrin beta 1(+)/CD200(+) VEGF(165) gene-modified iPS-HFSCsGFP combined with 3D printed degradable scaffolds containing 200 mu g/mL astragalus polysaccharide can promote early vascularization, collagen and hair follicle regeneration, and accelerate wound repair, achieving more complete structural and functional skin regenerative repair.

Summary: Vitiligo is a common chronic skin disease that negatively impacts patients' lives. Its pathogenesis involves autoimmunity and oxidative stress. Treatment should aim to control immune responses and promote melanocyte repigmentation. The Wnt/beta-catenin signalling pathway has emerged as a promising target for vitiligo therapy.
