Summary: This study demonstrates that DNA damage resulting from lagging chromosomes during mitosis and aneuploidy-induced replicative stress contributes to the increased invasiveness associated with chromosomal instability. The study also highlights the role of effector caspases and the JAK/STAT signaling pathway in enhancing DNA damage and promoting CIN-induced invasiveness. Additionally, an autocrine feedforward amplification loop mediated by Upd3 and the JAK/STAT signaling pathway is found to contribute to the activation of the apoptotic pathway in migrating cells, further promoting CIN-induced invasiveness.

Summary: This study conducted phytochemical profiling of Barleria hochstetteri using GC-MS and LC-MS methods, and investigated the cytotoxic effect of its extracts against breast and lung cancer cell lines using flow cytometry. The results revealed the presence of phenolic compounds and flavonoids with known cytotoxicity in the extracts, and demonstrated the cytotoxicity of the methanol extract on breast and lung cancer cells. Further research on B. hochstetteri is needed to uncover its potential as a source of anti-cancer drugs.

Summary: Inflammation promotes ovarian cancer development, and chemoresistance is a major challenge in its treatment. A series of gold(I) complexes derived from NSAIDs were synthesized, and complex B3 (Npx-Au) exhibited higher antitumor activity than cisplatin and other gold(I) complexes. Npx-Au induced oxidative stress and the DAMPs process by inhibiting TrxR activity. Mechanistic studies showed simultaneous downregulation of COX-2 and PD-L1 after Npx-Au treatment. Interestingly, in vivo experiments demonstrated that Npx-Au treatment stimulated immune response by reducing PD-L1 expression, inducing DC maturation, and increasing T cell infiltration. Overall, Npx-Au elicited immunogenic cell death and showed promise as a strategy for chemotherapy combined with immunotherapy in treating ovarian cancer.

Summary: During cell division, the activation of thousands of DNA replication origins ensures the complete duplication of the human genome. However, replication fork progression can be hindered by transcription-replication conflicts, with head-on conflicts being more detrimental. In this study, a bioinformatics toolkit called OKseqHMM was developed to analyze Okazaki fragment sequencing data and accurately predict replication initiation and termination, as well as replication fork directionality, in various organisms. This toolkit can greatly contribute to studying genome instability and its impact on human health.

Summary: This study found that HJURP is overexpressed in ovarian cancer and is associated with unfavorable prognosis. Knockdown of HJURP inhibits proliferation, metastasis, and induces cell stagnation in ovarian cancer cells. Mechanistically, HJURP regulates WEE1 through the MYC pathway, affecting chemosensitivity and DNA repair in ovarian cancer cells. Moreover, silencing HJURP enhances sensitivity to cisplatin and improves the synergistic effect of combined therapy with cisplatin and AZD1775 in ovarian cancer cells.
