Summary: In recent years, ambient ionization mass spectrometry (AIMS) has allowed for direct analysis of biofluid metabolome. However, AIMS procedures are hindered by analytical and practical limitations, which affect metabolome coverage. This study aimed to develop biofluid-specific metabolome sampling membranes (MetaSAMPs) to improve AIMS. Customized MetaSAMPs made of hydrophilic and lipophilic polymers demonstrated superior metabolome coverage and transport stability compared to crude biofluid analysis. This new approach was successfully validated and has potential applications in metabolic health stratification.

Summary: Plasma-based ambient desorption/ionization sources often use helium as the primary discharge gas, but studies show advantages of mixed-gas plasmas. Adding H-2 to helium can decrease reagent-ion signals while preserving analyte-ion signals. Nitrogen addition increases negative reagent ions for enhanced ion signal of specific compounds. Various molecular gases impact emission characteristics and plasma parameters in mixed-gas systems.

Summary: In this study, plasma chemistry in a helium flowing atmospheric pressure afterglow (FAPA) for analytical spectrometry is described using a quasi-one-dimensional plasma chemical kinetics model. The model demonstrates the formation of H, N, and O atoms in the discharge region, and the dominance of O-3 and NO species in the afterglow. Helium plays a key role in Penning ionization and charge transfer reactions, leading to high analytical sensitivity. The model's predictions align with previous observations in the literature, providing insights to enhance analytical performance of ambient desorption/ionization sources.

Summary: Ambient desorption/ionization mass spectrometry (ADI-MS) is a rapid screening tool that allows the analysis of samples without sample preparation. However, quantitative analysis of analytes in complex matrices remains a challenge. In this study, a carefully selected surface and co-deposited isotopically labeled standards were found to significantly improve the capabilities of FAPA-HR-MS for direct quantitative analysis. The use of TLC-based surfaces as sample carriers was shown to be an attractive tool for fast and efficient mass spectrometric investigations of complex samples.

Summary: An analytical platform combining probe sampling, thermal desorption-electrospray ionization/mass spectrometry (TD-ESI/MS), and multivariate statistical analysis was developed to rapidly differentiate bacterial species based on the differences in their lipid profiles. The results were validated by protein fingerprinting using matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF). The developed platform showed promising potential as a useful diagnostic tool for rapidly distinguishing bacterial species.

Summary: This study directly compared water-assisted laser desorption ionization (WALDI) and matrix-assisted laser desorption ionization (MALDI) mass spectrometry imaging, with MALDI as the benchmark for label-free molecular tissue analysis in biomedical research. The results showed that the two techniques are very similar in lipidomic analysis, especially when using norharmane MALDI matrix in negative ion mode. This similarity allows for the integration of data from both techniques to increase dataset and improve classification models.

Summary: In this study, liquid chromatography-high-resolution mass spectrometry methods were used to comprehensively characterize and quantify thymalfasin-related impurities. The results showed that there are various impurities in thymalfasin standard and commercial materials, with over half of them related to the C-terminal Asn residue. Therefore, it is proposed to modify or replace the C-terminal Asn residue of thymalfasin for new drug research and development.
