This study is to investigate thermal protection and moisture transmission of firefighter clothing and how combinations of materials influence the thermal protection and moisture transmission. An orthogonal design is applied for planning the experiment. Eight different material combinations, which simulate three-layer firefighter protective clothing system, are studied. The thermal protective performance (TPP) and water vapor transmission rate of combinations are tested. The experiment data are discussed by analysis of range and variance. Meanwhile, influence of each layer on clothing performance is also studied. Results show that outer shell fabric and moisture barrier layer have significant effects on TPP and moisture transmission property, respectively.

Regulatory CD4+and CD8+T cells are negatively correlated with CD4+/CD8+T cell ratios in patients acutely infected with SARS-CoV-2

Menglu Gao, Yili Liu, Mingquan Guo, Qianying Wang, Yan Wang, Jian Fan, Yinzhong Shen, Junjie Hou, Yanmin Wan, Zhaoqin Zhu

Summary: The study found that frequencies of lymphocytes and T cells decreased significantly in COVID-19 patients, while the CD4(+)/CD8(+)T cell ratios increased significantly in patients over 45 years old. Regulatory CD4(+) and CD8(+) T cells may play distinct roles in the pathogenesis of COVID-19, and NK cells might contribute to the lymphopenia associated with COVID-19.

An inter-correlated cytokine network identified at the center of cytokine storm predicted COVID-19 prognosis

Yili Liu, Daihong Chen, Junjie Hou, Haicong Li, Dan Cao, Mingquan Guo, Yun Ling, Menglu Gao, Yi Zhou, Yanmin Wan, Zhaoqin Zhu

Summary: Elevated plasma levels of IL-6, IL-8, and IL-5 were consistently found in deceased COVID-19 patients compared to those with mild/moderate or severe disease. The early plasma concentrations of IFN-a and IL-2 positively correlated with the length of the disease course. IL-6, IL-8, and IL-5 may play central roles in cytokine storms associated with COVID-19.

Exploration of a Sequential Gp140-Gp145 Immunization Regimen with Heterologous Envs to Induce a Protective Cross-Reactive HIV Neutralizing Antibody Response In Non-human Primates

Xiangqing Ding, Kangli Cao, Jing Wang, Yanmin Wan, Qinyun Chen, Yanqin Ren, Yongtang Zheng, Mingzhao Zhu, Renrong Tian, Wenjun Wang, Chen Zhao, Xiaoyan Zhang, Jianqing Xu

Summary: The study explored the utility of diverse HIV-1 envelope immunogens in a sequential immunization scheme to raise a heterologous tier 2 nAb response for HIV vaccine development. By implementing a three-immunization regimen in rhesus macaque over a short 8-week schedule, a potent nAb response with substantial breadth was elicited, effectively containing viremia upon a heterologous SHIV challenge. This highlights the potential of diversifying Env immunogens under a sequential immunization scheme for HIV vaccine development.

Summary: Researchers found that T follicular helper (T-FH) cells are highly sensitive to ferroptosis, and this process is regulated by the activity of the selenoenzyme GPX4. Selenium supplementation can increase GPX4 expression, leading to an increase in T-FH cell numbers and antibody responses.

Summary: Leptin levels are associated with reduced vaccine responses, as it promotes T-FH differentiation and IL-21 production. Leptin deficiency impairs T-FH generation and antibody responses, increasing the risk of vaccine failure.

Omicron variant showed lower neutralizing sensitivity than other SARS-CoV-2 variants to immune sera elicited by vaccines after boost

Summary: This study explored the immunogenicity of different vaccination strategies against the Omicron variant of SARS-CoV-2. The results showed that the Omicron variant has a high immune escape ability compared to other variants, but heterologous protein subunit vaccines and homologous inactivated vaccine boosters can improve neutralization against Omicron.

An Update on the Mutual Impact between SARS-CoV-2 Infection and Gut Microbiota

Shaoshuai Li, Yang Zhou, Dongmei Yan, Yanmin Wan

Summary: The gut microbiota and COVID-19 have a mutual impact on each other. COVID-19 can affect the composition of the gut microbiota, and changes in the gut microbiota may be related to the severity of COVID-19. Additionally, there is immune cross-reactivity between SARS-CoV-2 and commensal bacteria.

Immune Correlates of Disseminated BCG Infection in IL12RB1-Deficient Mice

Xuyang Wang, Liqiu Jia, Yang Liu, Jing Wang, Chao Qiu, Tao Li, Wenhong Zhang, Zhaoqin Zhu, Jing Wu, Yanmin Wan

Summary: Individuals with IL12RB1 deficiency are more susceptible to BCG and environmental mycobacteria infections. This study using a mouse model reveals that IL12RB1-deficient mice have impaired tuberculosis suppression, with decreased innate immune responses and increased IL-17 responses. It suggests that IFN-gamma responses alone may not be sufficient to contain BCG infection in the setting of IL12RB1 deficiency.

Unexpectedly higher levels of anti-orthopoxvirus neutralizing antibodies are observed among gay men than general adult population

Summary: This study compared the antibody responses against monkeypox virus (MPXV) and vaccinia virus in cohorts of gay men and the general adult population. It found that individuals born in/after 1981 in the gay men cohort had lower antibody responses against MPXV but higher neutralizing antibody responses against vaccinia compared to the general population. These findings provide insights into the impact of preexisting antibodies on MPXV transmission in the MSM community.

Summary: In this study, it was found that the inactivated COVID-19 vaccine was safe and immunogenic in HIV-1 infected individuals who are stable on cART. Participants did not experience significant adverse reactions after vaccination. Interestingly, HIV-infected individuals with low baseline CD4(+)/CD8(+) T cell ratios exhibited lower antibody responses post-vaccination compared to those with higher ratios.

High levels of soluble CD25 in COVID-19 severity suggest a divergence between anti-viral and pro-inflammatory T-cell responses

Min Xie, Joseph Yunis, Yin Yao, Jing Shi, Yang Yang, Pengcheng Zhou, Kaili Liang, Yanmin Wan, Ahmed Mehdi, Zhian Chen, Naiqi Wang, Shuyun Xu, Min Zhou, Muqing Yu, Ke Wang, Yu Tao, Ying Zhou, Xiaochen Li, Xiansheng Liu, Xiao Yu, Yunbo Wei, Zheng Liu, Jonathan Sprent, Di Yu

Summary: The study aimed to understand the immune response in COVID-19 patients with T cells showing both functional defects and hyperactivation. Elevated levels of sCD25 were found in severe cases, indicating insufficient anti-viral immunity and expansion of pro-inflammatory T cells. The mouse model and clinical observation confirmed the association between high sCD25 levels and disease severity.
