Summary: In this study, the inhibitory potential of four major catechins (EC/EGC/ECG/EGCG) against CCL2-regulated monocyte/macrophage recruitment was assessed. Catechins with the gallate moiety (ECG/EGCG) specifically attenuated CCL2-induced macrophage migration by binding to a pocket on CCL2. The molecular specificity/stability of CCL2-catechin complexes was regulated by multiple factors, including stereospecificity, hydroxyl groups, carbonyl group positioning, and galloylation.

Summary: This study found that an intervention using l-theanine (LTA) and epigallocatechin gallate (EGCG) can alleviate ovalbumin allergy (OVA-A) by modulating intestinal-specific immunity. The combination of LTA and EGCG has a better effect than using them separately. Moreover, LTA and EGCG also improve OVA-A by reducing the number of dendritic cells, inhibiting the Th2 immune response, and inhibiting mast cell degranulation.

CGA plus EGCG improved cognition deficits and protected gut barrier function more effectively compared to the agents alone. Additionally, CGA and EGCG modulated the gut microbiome and reduced gut inflammation.

Summary: This study investigated the effects of EGCG on interface dermatitis (ID) by analyzing its impact on the expression of interferon-associated mediators CXCL10 and MxA in both lesional skin of lichen planus patients and epidermal equivalents. The results showed that EGCG can reduce the intensity of CXCL10 and MxA staining in epidermal equivalents and secretion of CXCL10 by keratinocytes, suggesting its potential as a topical therapeutic in autoimmune skin diseases associated with ID.

Summary: Cardiac tissue damage following ischemia leads to cardiomyocyte apoptosis and myocardial fibrosis. The active polyphenol flavonoid or catechin epigallocatechin-3-gallate (EGCG) protects ischemic myocardium and is involved in the regulation of endothelial-to-mesenchymal transition (EndMT). In this study, EGCG treatment was shown to regenerate ischemic tissue and induce cardioprotection by regulating proteins associated with the EndMT process, as well as positively regulating cardiomyocyte apoptosis and fibrosis.

Summary: Chronic neuroinflammation is associated with neurodegenerative diseases and activated microglia cells play a crucial role in this process. Epigallocatechin 3-gallate (EGCG) is a natural compound with anti-inflammatory and antioxidant properties that shows promise in treating inflammation-related neurodegenerative diseases. This study aimed to explore the molecular targets of EGCG in activated microglia cells and found that EGCG effectively inhibited proinflammatory mediator NO production and downregulated MIF, CCL-2, and CSF2. Additionally, EGCG modulated multiple signaling pathways involved in neuroinflammation, suggesting its potential as a therapeutic agent for neurodegenerative diseases.

Summary: This study aims to illustrate the in vitro and in vivo protective mechanism of epigallocatechin gallate (EGCG) on SEA-triggered intestinal barrier damage and inflammation. Results show that EGCG alleviates intestinal barrier damage by effectively inhibiting SEA-induced intestinal permeability increase, tight junction protein and mucin loss, and intestinal cell apoptosis. EGCG also reduces intestinal inflammation by suppressing the TLR4-NF-kappa B/MAPKs-NLRP3 pathway. Importantly, EGCG reverses gut microbiota dysbiosis and short-chain fatty acid (SCFA) content decrease induced by SEA.

Adenovirus-mediated over-expression of IL-1, IL-23, and IL-7 can rescue mice from Pseudomonas aeruginosa-induced lethality, suggesting its potential value in locally immunosuppressed individuals like cystic fibrosis patients.

Summary: Increasing evidence suggests that SARS-CoV-2 infection affects neurological function, and developing laboratory mouse models is important for studying the underlying mechanisms. Our study demonstrates that SARS-CoV-2 (MA10) infection induces neuroinflammation in laboratory mice and could be used as a novel model to study SARS-CoV-2-mediated cerebrovascular pathology.
