Strengths and weaknesses of alternative noninvasive imaging approaches for microvascular ischemia

Isabella Leo, Eleni Nakou, Jessica Artico, Emmanouil Androulakis, Joyce Wong, James C. Moon, Ciro Indolfi, Chiara Bucciarelli-Ducci

Summary: Structural and functional abnormalities of coronary microvasculature are common in clinical settings and associated with worse clinical outcomes. Traditional invasive methods for assessment of coronary microvascular function have limitations and risks. Recent advances in non-invasive imaging techniques provide alternative approaches for evaluating coronary microvascular function.

Resident memory T cells develop regional dialects

Nicholas J. Maurice, Stephen C. Jameson

Summary: Human resident memory T cells segregate in different tissues and have gene expression patterns specific to those tissues, suggesting the acquisition of site-specific immunity beyond shared residency traits.

Toward a general model of CD4+T cell subset specification and memory cell formation

Kevin C. Osum, Marc K. Jenkins

Summary: In the field of CD4+ T cells, there has been a lack of progress compared to the transformative discoveries made in memory CD8+ T cell biology in recent decades. This perspective focuses on the specification of CD4+ helper T (Th) cell subsets and the formation of memory cells. The authors argue that the abundance of Th effector and memory cell subsets and the emphasis on their differences have hindered the development of a general model for CD4+ memory T cell formation applicable to all immune responses. They propose a bifurcation model based on an IL-2 signal-dependent switch to explain the balanced production of diverse Th memory cells involved in cell-mediated or humoral immunity.

Detection of subclinical hypertrophic cardiomyopathy

George Joy, James C. Moon, Luis R. Lopes

Summary: Hypertrophic cardiomyopathy (HCM) is characterized by ventricular hypertrophy, but it also involves other abnormalities such as cardiomyocyte disarray, myocardial ischemia, and electrical issues, which may appear before the onset of hypertrophy. With advancements in genetic testing and therapeutic agents, detecting subclinical HCM has become a growing priority. In this context, novel biomarkers like quantitative perfusion and diffusion tensor MRI play a significant role.

Persistent symptoms after COVID-19 are not associated with differential SARS-CoV-2 antibody or T cell immunity

Daniel M. Altmann, Catherine J. Reynolds, George Joy, Ashley D. Otter, Joseph M. Gibbons, Corinna Pade, Leo Swadling, Mala K. Maini, Tim Brooks, Amanda Semper, Aine Mcknight, Mahdad Noursadeghi, Charlotte Manisty, Thomas A. Treibel, James C. Moon, Rosemary J. Boyton

Summary: This study analyzes the immune response in healthcare workers with mild or asymptomatic SARS-CoV-2 infection and compares it to those with Long Covid. The results show no significant differences in immune responses between the two groups up to 18 weeks post-infection and 1 year after vaccination, suggesting that immune response variations are unlikely to be a contributing factor to Long Covid development.

Intestinal epithelial HDAC3 and MHC class II coordinate microbiota-specific immunity

Emily M. Eshleman, Tzu-Yu Shao, Vivienne Woo, Taylor Rice, Laura Engleman, Bailey J. Didriksen, Jordan Whitt, David B. Haslam, Sing Sing Way, Theresa Alenghat

Summary: Abnormal immune responses can cause inflammatory bowel disease and other chronic inflammatory conditions. This study found that mice lacking the expression of histone deacetylase 3 (HDAC3) in the intestinal epithelium had increased accumulation of commensal-specific CD4+ T cells, suggesting that HDAC3 may control local microbiota-specific immunity. Further experiments showed that HDAC3 was necessary for regulating epithelial MHC class II (MHCII) and dampening the accumulation of commensal-specific Th17 cells, protecting against microbiota-triggered inflammation. These findings highlight the important role of HDAC3 in regulating the balance of tissue-specific CD4+ T cell subsets and controlling inflammation.
