Triazole linker-based trivalent sialic acid inhibitors of adenovirus type 37 infection of human corneal epithelial cells

Adenovirus type 37 (Ad37) is one of the principal agents responsible for epidemic keratoconjunctivitis (EKC), a severe ocular infection that remains without any available treatment. Recently, a trivalent sialic acid derivative (ME0322, Angew. Chem. Int. Ed., 2011, 50, 6519) was shown to function as a highly potent inhibitor of Ad37, efficiently preventing the attachment of the virion to the host cells and subsequent infection. Here, new trivalent sialic acid derivatives were designed, synthesized and their inhibitory properties against Ad37 infection of the human corneal epithelial cells were investigated. In comparison to ME0322, the best compound (17a) was found to be over three orders of magnitude more potent in a cell-attachment assay (IC50 = 1.4 nM) and about 140 times more potent in a cell-infection assay (IC50 = 2.9nM). X-ray crystallographic analysis demonstrated a trivalent binding mode of all compounds to the Ad37 fiber knob. For the most potent compound ophthalmic toxicity in rabbits was investigated and it was concluded that repeated eye administration did not cause any adverse effects.

Synthesis of 4-O-Alkylated N-Acetylneuraminic Acid Derivatives

Summary: This paper describes the synthesis of 4-O-alkyl analogues of N-acetylneuraminic acid (Neu5Ac) and the scope of the reaction. It demonstrates that activated alkyl halides and sulfonates, as well as primary alkyl iodides, can give products in useful yields. These results expand the toolbox of Neu5Ac chemistry and have value in drug discovery and the study of Neu5Ac lectins biology.

Ex Vivo and In Vivo CD46 Receptor Utilization by Species D Human Adenovirus Serotype 26 (HAdV26)

Summary: This study investigates the usage of CD46 receptor by Ad26 virus in different administration routes and its impact on infection efficiency. The results show that Ad26 can use CD46 as a receptor but its usage varies depending on the administration route. Ectopic expression of CD46 on cells enhances Ad26 infection, while tumor injection or intravenous injection shows lower infection efficiency. High-dose intravenous injection of Ad26 induces liver enzyme release and reduces white blood cell counts but does not cause thrombocytopenia.

Various methods have been established for regenerating expensive cofactors to make the enzymatic process more efficient and economically viable.
