Summary: This study investigated the prevalence and associations of p16 positivity with survival in HNSCC patients in Southwest Finland between 2005 and 2015. The results showed that p16 positivity was mainly found in the oropharynx and was associated with an increased likelihood of lymph node metastasis, a smaller primary tumor size, and a sparse history of smoking and alcohol consumption. Regardless of p16 expression, all HNSCC patients benefited from combining chemotherapy and radiotherapy.

The prevalence of p16 positivity and double positivity increased over time, and nonkeratinizing and warty/basaloid VSCCs had higher double positivity rates.

The results showed synergistic effects of BMN673 and MK-1775 on SHH cell lines, but not on group 3 cell lines. MK-1775 alone may be of interest for all MB cell lines, and the combination of PARP/WEE1 inhibitors may provide possible therapeutic opportunities for SHH MBs.

Summary: Adenoid cystic carcinoma (AdCC) is a rare heterogeneous disease that is difficult to diagnose and predict prognosis, making treatment challenging. A retrospective study on a large cohort of AdCC patients in the head and neck revealed that early disease stage and major salivary gland subsite were favorable prognostic factors, while perineural invasion and radical surgery did not significantly correlate with survival. These findings have important implications for prognostic assessment of AdCC.

Summary: In order to identify patients with HPV-positive OPSCC and a poor prognosis after treatment, the gene expression in tumours from patients with poor or favourable prognosis was compared in a case-control study. The results were validated on RNA and protein levels in two separate cohorts. High expression of FGF11 was associated with poor patient survival in all three cohorts. This suggests that FGF11 may be a major prognostic marker in HPV-positive OPSCC.

The results provide the first comprehensive molecular atlas for primary GCOC, including previously identified CTNNB1 mutation and novel alterations in MAP3K, EP300, and the 22q11.21 region. The transcriptome analysis reveals significant involvement of cytokine-cytokine receptor interaction and the PI3K-Akt signaling pathway. More GCOC cases should be compared to validate these findings for accurate clinical guidance.

Summary: This study compares the outcomes of transoral resections using the da Vinci SP and da Vinci Xi systems for oropharyngeal squamous cell carcinoma (OPSCC). The results indicate that the da Vinci SP system has advantages in terms of reduced docking and console times, as well as improved rates of wide negative resection margins. However, there were no significant differences in complication rates or postoperative functional outcomes between the two systems.
