Summary: NASH is an important stage of NAFLD in humans, and there are currently no approved drugs for its treatment. The upregulation of AKR1B10 gene in NASH patients has been identified, and the inhibition of AKR1B10 has shown promising effects in reducing the pathological features of NASH.

Summary: This study found that corn oligopeptides (CPs) have a regulatory effect on alcohol-induced liver injury and inflammation. CPs can reduce the levels of inflammatory cytokines and liver function indicators, and inhibit the activation of inflammatory signaling pathways and the expression of inflammatory cytokines. These findings suggest that CPs have potential application value in the treatment of acute alcoholic liver disease.

Summary: This pilot study evaluated the potential of the Nod-like receptor (NLR) signaling pathway related RNA panel in diagnosing non-alcoholic steatohepatitis (NASH). Upregulation of Lnc-SPARCL1-1:2 lncRNA led to increased levels of miR-6881-5P and subsequent mRNA levels of HSPD1, MMP14, and ITGB1 were observed. ROC curve analysis showed discriminative cutoff values that aided in distinguishing NASH cases from controls, as well as differentiating between NAFLD, simple steatosis, and NASH.

Summary: The study revealed that hypoxia exposure can promote the development of NASH by upregulating HIF-2 alpha, further activating NF-kappa B, exacerbating fibrosis and inflammation, providing a new approach for the treatment of NASH.

Summary: Corosolic acid (CA) from hawthorn has shown significant therapeutic effects on non-alcoholic steatohepatitis (NASH), ameliorating liver inflammation and fibrosis by regulating TGF-beta 1/Smad2, NF-kappa B, and AMPK signaling pathways.

Summary: This study evaluated the safety and efficacy of combining semaglutide with cilofexor and/or firsocostat in patients with NASH. The combination therapy was generally well tolerated and resulted in additional benefits over semaglutide monotherapy, including improvements in liver steatosis and biochemistry. Double-blind placebo-controlled trials with larger patient numbers are needed to further assess the efficacy and safety of these combination therapies in NASH.

Summary: Medulloblastoma is a common malignant pediatric brain tumor, and metastasis and relapse are the leading causes of death in patients. This study reveals that AMPK inhibits the proliferation and metastasis of SHH-MB cells through the SHH-GLI1 and NF-kappa B signaling pathways. Blocking the NF-kappa B and Shh pathways together synergistically restrains the growth and invasion of SHH-MB cells, suggesting a potential molecular therapy target.

Summary: In this study, a network pharmacology approach was used to identify the TLR4 signaling pathway as a primary target of hesperitin for the treatment of alcoholic steatohepatitis (ASH). Molecular docking results showed that hesperitin bound to TLR4 and exhibited good affinity. Furthermore, hesperitin inhibited the TLR4 target and consequently the NF-kappa B signaling pathway, which slowed the evolution of alcoholic steatohepatitis.

Summary: Resmetirom, an orally active agonist of THR-beta that targets the liver, shows promise in treating NASH, but its underlying mechanism is not well understood. This study established a NASH cell model to test the preventive effect of resmetirom and used RNA-seq for screening. They also conducted rescue experiments to validate the drug's target gene. A NASH mouse model was used to further investigate the role and mechanism of resmetirom. The findings showed that resmetirom effectively reduced lipid accumulation and triglyceride levels and recovered the expression of RGS5, which is repressed in the NASH model.

Summary: Cholesterol sulfate (CS) activates retinoic acid-related orphan receptor a (ROR alpha). CS and ROR alpha inhibit osteoclast differentiation. The mechanism involves the AMPK-Sirt1 axis, leading to NF-kappa B inhibition. CS also induces apoptosis and protects against bone loss in mouse models.

Summary: The study identified 381 lncRNAs highly expressed in NAFLD and even higher in NASH. A novel LncRNA-gm9795 was found to be significantly upregulated in NASH animal and cell models. The research indicated that gm9795 promotes inflammatory response in NASH through NF-kB and JNK pathways.

Summary: The PNPLA3 and TM6SF2 variants are associated with non-alcoholic fatty liver disease (NAFLD) and its progression. This study aimed to evaluate the association between genetic ancestry, represented by ancestry informative markers (AIM), and PNPLA3 and TM6SF2 polymorphisms in patients with biopsy-proven NAFLD in an admixed population. The results showed that greater African genetic ancestry was associated with a lower frequency of the PNPLA3 G allele, indicating a potential protective factor against NAFLD in individuals with African ancestry.

Summary: This study examined the effect of IMI exposure on autophagy in fish kidney cells and the potential of eucalyptol to protect against the toxicity of IMI. The results showed that IMI induced autophagy in the cells, while eucalyptol treatment alleviated the autophagy and its associated signals. These findings provide insights into the mechanism of biological threat posed by IMI exposure to fish and the potential application of eucalyptol in aquaculture.
