Summary: The study found that alternative lipid sources rich in saturated and monounsaturated fatty acids can be used in cobia aquafeeds when supplemented with long-chain polyunsaturated fatty acids without impairing production performance. High levels of saturated fatty acids were disproportionally deposited in the liver and muscle, while monounsaturated fatty acids were mainly deposited in the liver and muscle.

Summary: Fatty acids are an important energy source and component of cell membranes. This study investigates the impact of the balance between saturated fatty acids (SFAs) and unsaturated fatty acids (UFAs), regulated by stearoyl Co-A desaturase (SCD), on ovarian cancer cell survival and tumor progression. Depletion or inhibition of SCD leads to lower levels of UFAs, triggering endoplasmic reticulum (ER) stress response and cell death. However, supplementation with UFAs and reequilibration of SFA/UFA levels rescues stress and apoptosis. This research highlights the importance of targeting lipid balance in the treatment of ovarian cancer.

Summary: We investigated the role of ER-phagy in NAFLD-related hepatocarcinogenesis in HFD-fed and/or PB-treated rats by clustering the expression levels of p62 and FAM134B in preneoplastic hepatic lesions. The results showed that PB administration increased clusters with high levels of p62, while HFD feeding increased clusters with high levels of both p62 and FAM134B. The combination of HFD feeding with PB counteracted each other's effects and the cluster composition resembled that of the control group.

Summary: Metabolic syndrome affects a large number of individuals worldwide and is a major cause of health problems. Obesity is at the core of metabolic abnormalities associated with MetS. This study found that a metabolic enhancer composed of seven naturally occurring antioxidants and mitochondrial enhancing agents improved obesity, hepatic steatosis, and atherogenic serum profile in mice. The findings suggest that the components of the metabolic enhancer have a positive, protective effect on obesity, hepatic steatosis, and cardiovascular risk similar to exercise training.

Summary: Non-alcoholic fatty liver disease (NAFLD) affects one billion individuals worldwide and is caused by increased intake of a high-fat diet (HFD) and sugar-sweetened beverages. The study reveals that fructose metabolism via ketohexokinase (KHK) C isoform leads to unresolved endoplasmic reticulum (ER) stress in combination with a HFD intake, and a liver-specific knockdown of KHK can improve NAFLD activity. Additionally, upregulation of KHK-C is associated with obesity, insulin resistance, and liver triglycerides in both mice and humans.

Summary: Ferroptosis, an iron-dependent cell death, is observed in the development of non-alcoholic fatty liver disease (NAFLD). Melatonin shows potential benefits in preventing and treating liver diseases. In this study, a mouse model of NAFLD induced by long-term high-fat diet feeding was established, and it was found that Melatonin treatment improved metabolic abnormalities and inhibited the progression of NAFLD. Melatonin supplementation also improved iron homeostasis disorders and hepatic lipid peroxidation induced by high-fat diet. Mechanistically, Melatonin inhibited ER stress through the MT2/cAMP/PKA/IRE1 pathway to inhibit hepatic ferroptosis.

Summary: Sar1, a small GTPase protein of the ARF family, plays a role in cargo concentration and membrane deformation by associating with the endoplasmic reticulum membrane and recruiting COPII components. The regulation of Sar1's GTP hydrolysis and its role in coat assembly, as well as the mechanism of Sar1-induced membrane deformation and scission, are still not fully understood, especially in higher organisms with a wider range of secreted cargoes. This review focuses on these aspects and highlights the additional specialization in higher eukaryotes, along with the outstanding questions regarding the orchestration of Sar1 functions.
