Summary: The NGS mutational analysis of bile cfDNA, named Bilemut, showed a sensitivity of 96.4% and specificity of 69.2% for detecting malignancy in patients with suspicious biliary strictures. Implementation of Bilemut at the initial diagnostic stage can significantly improve detection of malignancy, reduce delays in clinical management, and assist in selecting patients for targeted therapies.

Summary: In this study, we aimed to identify BTC-specific DNA methylation alterations. By performing a genome-wide methylation study and validating the findings in multiple samples, we successfully identified and validated BTC-specific DNA methylation alterations in over 200 BTC samples. Additionally, we discovered a biomarker associated with the HOXD8 gene that showed high sensitivity and specificity in detecting BTC in tissue and bile samples.

Summary: The tumor suppressive function of CYLD has been identified in melanoma, but the precise molecular mechanism remains unclear. This study generated a novel melanoma mouse model and found that CYLD plays a role in chromatin structure formation through histone methylation. These findings reveal a new role of CYLD in melanoma progression.

Summary: Systemic lupus erythematosus (SLE) is a common autoimmune disorder, with lupus nephritis (LN) being a frequent and difficult to treat complication. This study identified the multifunctional cytokine AREG as an anti-inflammatory mediator in LN, suppressing pathogenic T cell immunity. These findings highlight the potential of the AREG/EGFR axis as a therapeutic target.

Summary: By performing proteomic and metabolomic analyses, we identified new pro-tumorigenic mechanisms in intrahepatic cholangiocarcinoma (iCCA), including activation of EGFR signaling or KRAS mutation driving IL6 expression in tumor cells, as well as reprogramming of glucose metabolism and activation of the serine-glycine pathway. These findings provide potential therapeutic targets for iCCA.

Summary: This study identifies a set of lncRNAs that are significantly downregulated in hepatocellular carcinoma (HCC) and are related to tumor dedifferentiation and patients' prognosis. Mechanistically, these lncRNAs are involved in hepatic differentiation and their expression is silenced by DNA methylation in HCC. These findings provide insights into the progression of liver cancer and potential new biomarkers.

Loss of liver function in chronic liver disease: An identity crisis

Summary: The identity of adult hepatocytes is established during embryonic development and regulated after birth. Transcription factors and pre-mRNA splicing regulators play a crucial role in defining the transcriptome, which determines the complex functions of the liver. Persistent downregulation of key identity genes is a major factor in chronic liver disease and organ dysfunction. Therefore, understanding the disruption of core transcription factors and splicing regulators in diseased hepatocytes is clinically important.
