Inhibition of Matrix Metalloproteinase Activities and Tightening of Tight Junctions by Diallyl Disulfide in AGS Human Gastric Carcinoma Cells

The effect of diallyl disulfide (DADS), a major component of an oil-soluble allyl sulfide garlic (Allium sativum) derivative, on the correlation between anti-invasive activity and tightening of tight junctions (TJs) was investigated in human gastric adenocarcinoma AGS cells. Our data indicated that the inhibitory effects of DADS on cell motility and invasiveness were found to be associated with increased tightness of the TJs, which was demonstrated by an increase in transepithelial electrical resistance. Activities of matrix metalloprotease (MMP)-2 and -9 in AGS cells were dose-dependently inhibited by treatment with DADS, and this was also correlated with a decrease in expression of their mRNA and proteins; however, tissue inhibitor of metalloproteinase (TIMP)-1 and -2 mRNA levels and proteins were increased. Additionally, immunoblotting results indicated that DADS repressed the levels of claudin proteins (claudin-2, -3, and -4), major components of TJs that play key roles in control and selectivity of paracellular transport. Although further studies are needed, these results suggest that DADS treatment may inhibit tumor cell motility and invasion and, therefore, act as a dietary source to decrease the risk of cancer metastasis.

This signature includes three distinct molecular subtypes, with DP.BCG+ related to progression and response to BCG treatment, and REC.BCG+ related to recurrence and response to BCG treatment. These findings enhance understanding of NMIBC prognosis and the effectiveness of BCG therapy, potentially improving clinical practice.

These findings suggest that loquat leaf extract may be a potential therapeutic candidate for treating myoblast injury and enhancing cellular differentiation.

Effects of Rosemary Extract on C2C12 Myoblast Differentiation and 5-Aminoimidazole-4-carboxamide Ribonucleoside (AICAR)-Induced Muscle Cell Atrophy

Summary: The effectiveness of rosemary extract (RE) in inducing myotube differentiation and inhibiting AICAR-induced muscle atrophy was evaluated in this study. The results showed that RE increased myotube thickness and promoted the expression of muscle differentiation-related proteins MyoD and myogenin. Furthermore, RE reduced the expression of muscle atrophy-related proteins FoxO3a, MAFbx/atrogin-1, and MuRF-1, while significantly increasing the expression of MyoD and myogenin.

Summary: In this study, the anti-fibrotic effects of derrone on TGF-beta 1-stimulated MRC-5 lung fibroblast cells and bleomycin-induced lung fibrosis were investigated. It was found that high concentrations of derrone increased cytotoxicity in MRC-5 cells, but low concentrations did not induce significant cell death. Derrone reduced the expression of TGF-beta 1, fibronectin, elastin, and collagen1 alpha 1 in TGF-beta 1-stimulated MRC-5 cells, and also attenuated fibrotic histopathological changes in bleomycin-treated mice.

Targeted Deletion of Thymosin Beta 4 in Hepatic Stellate Cells Ameliorates Liver Fibrosis in a Transgenic Mouse Model

Summary: Liver fibrosis is primarily caused by activated hepatic stellate cells (HSCs), and targeting these cells is crucial in preventing liver fibrosis. This study investigated the in vivo functions of thymosin beta 4 (T β 4) in liver fibrosis by generating T β 4 conditional knockout mice. The selective deletion of T β 4 in activated HSCs attenuated liver injury and fibrosis by repressing Hedgehog signaling. Re-expression of T β 4 reversed the effect of HSC-specific T β 4 deletion and led to liver fibrosis with Hedgehog activation. These findings suggest T β 4 as a potential therapeutic target for liver fibrosis.
