Summary: The study reveals the significance of high-order three-dimensional chromatin interactions in gene regulation. By developing Pore-C and Chromunity, researchers are able to analyze high-order chromatin contacts at the genome scale and find their enrichment in enhancers and promoters in active chromatin as well as highly transcribed and lineage-defining genes.

Summary: DNAJA4 is hypermethylated and downregulated in nasopharyngeal carcinoma (NPC). Overexpression of DNAJA4 inhibits NPC cell migration, invasion, and EMT by promoting MYH9 protein degradation. Low level of DNAJA4 is associated with poor prognosis and increased probability of distant metastasis in NPC patients.

Summary: This study investigated the methylation status of TSHR, PTEN, and RASSF1A genes in the plasma of patients with PTC. The results showed that the methylation level of TSHR was significantly higher in both the thyroid cancer tissue and plasma samples. Additionally, TSHR methylation level was associated with certain clinical characteristics of PTC and demonstrated high accuracy in distinguishing PTC from healthy controls.

Summary: The RAS gene, consisting of KRAS, HRAS, and NRAS, is associated with human tumors, with KRAS having the highest mutation incidence at around 80% of cases. This gene plays a regulatory role in transcription and translation at the molecular level, and affects cell proliferation and migration at the cellular level, making it crucial for cancer development. In 2021, the FDA approved the first direct inhibitor, AMG510, targeting the KRAS-G12C mutation, which showed tumor regression, prolonged survival, and low off-target activity. However, due to drug resistance, single inhibitors are no longer sufficient, leading to the development of numerous highly efficient inhibitors.

Summary: Saliva sampling is a non-invasive method that can be done by individuals themselves. This study used saliva samples to explore the use of EBV methylation as a biomarker in the diagnosis of NPC. The results showed that EBV methylation was significantly increased in NPC patients compared to controls, and detection of specific CpG sites showed good diagnostic performance. The study suggests that non-invasive detection of NPC through saliva sampling could be a promising alternative for future large-scale screening.

Summary: Viral oncogenes can modulate cellular metabolic reprogramming to drive malignant transformation of normal epithelia cells. Understanding the dysregulation of viral oncogene-mediated signaling transduction in metabolic reprogramming may offer new therapeutic targets for virus-associated cancer treatment. This review focuses on elucidating how EBV infection and its encoded oncoproteins alter metabolic reprogramming to promote malignancy, and provides a new perspective on the interplay between EBV infection and metabolic pathways, offering a potential therapeutic intervention strategy for EBV-associated malignant diseases.
