This article provides an overview of current methods for intracellular protein delivery, highlighting challenges and opportunities for future research.

Summary: Pulmonary delivery is a non-invasive approach for administering biotherapeutics. This study focuses on the receptor-mediated delivery of protein cargo using non-covalent protein complexes. The complexes achieve intracellular delivery via the biotin receptor and show preferential involvement of dynamin- and caveolae-dependent vesicular internalization. The study provides evidence of intracellular presence of the complexing copolymer, indicating the potential for protective intracellular delivery of biotherapeutics.

Summary: This study examined the correlation between albumin levels and inflammatory indices in hospitalized patients, showing negative correlations between albumin and CRP/white blood cell levels and positive correlations with platelets/hemoglobin levels. The strongest negative correlation between albumin and CRP was found in the Internal Medicine departments. Linear regression analysis revealed a minor effect of CRP on albumin levels, particularly at very high CRP levels. Further exploration of the mechanisms underlying this negative correlation is needed.

Summary: Protein-antibody conjugates (PACs) offer a promising approach for targeted delivery of protein therapeutics to specific cells by combining the advantages of biologics and antibody-drug conjugates. Utilizing spherical polymer brushes and orthogonal click chemistries, PACs can specifically target cells based on the presence of target antigen on the cell surface. The adaptability of PACs opens up new opportunities for targeted biologics in therapeutics and sensing.

This study investigated the use of ionic polyphosphazenes to deliver protein cargo intracellularly, finding that PZ-PYR showed faster internalization and higher uptake levels in endothelial cells compared to PZ-PEG, although both polymers achieved similar uptake levels in cancer cells. Uptake was mediated by endocytosis through multiple mechanisms.

Summary: SCP has emerged as a novel intracellular delivery system with enhanced efficiency and serum tolerance. However, its application is limited due to cationic charge and non-specificity. Researchers have engineered an activatable SCP (ASCP) with pH-sensitive charge shielding sequence, protease cleavage site, and SCP. This system reduces non-specific binding and delivers various cargo into the cytosol both in vitro and in vivo. The ASCP fusion protein enables co-delivery of therapeutic cargos for optimal outcomes and paves the way for combination cancer therapies.

Participants' education levels and baseline knowledge scores were closely associated with knowledge improvement in appropriate warfarin use.

In the first quarter of 2019, a total of 260 pharmacists were included in the analysis, with significant cost savings and effectiveness observed in pharmaceutical care, especially in ICUs.
