Topical drug delivery to the posterior segment of the eye: The effect of benzalkonium chloride on topical dexamethasone penetration into the eye in vivo

Benzalkonium chloride (BAC) is a quaternary ammonium compound with antibacterial properties. It is used as preservative in aqueous eye drops but studies have shown that it can also act as a penetration enhancer increasing drug permeation from the eye surface into the eye. Here the effect of BAC on dexamethasone permeation into the eye was investigated in vivo. Dexamethasone in aqueous cyclodextrin-based eye drops with and without BAC, were administered to rabbits and the dexamethasone concentration in several eye tissues determined 2 h after the administration. The study shows that BAC has no effect on the drug penetration from the eye surface into the eye. The penetration enhancing effect of BAC is mainly based on in vitro observations where BAC is in contact with isolated epithelial membrane (e.g., cornea) for several hours. Under in vivo conditions, BAC is rapidly washed from the eye surface with the tear fluid before it can penetrate into the epithelia and disrupt its structure sufficiently to allow for enhanced drug permeation into the eye.

Effect of salt formation on γ-cyclodextrin solubilization of irbesartan and candesartan and the chemical stability of their ternary complexes

Summary: This study aimed to increase the aqueous solubility of angiotensin II receptor blockers (ARBs) by forming cyclodextrin inclusion complexes and determine their chemical stability in aqueous solutions. The results showed that the solubility of drugs increased by forming inclusion complexes with gamma-cyclodextrin and by increasing their intrinsic solubility in phosphate buffer. The formation of inclusion complexes was confirmed by molecular docking. Salt formation using various organic salts enhanced the solubilization of drugs with gamma-cyclodextrin, but chemical instability was observed at high salt concentrations.

Cyclodextrin-based Pickering nanoemulsions containing amphotericin B: Part I. evaluation of oil/cyclodextrin and amphotericin B/cyclodextrin inclusion complexes

Summary: The solubility of amphotericin B (AmB) and the effect of cyclodextrins (CDs) on the complex formation of medium chain triglyceride (MCT) oils and AmB were investigated prior to the development of CD stabilized Pickering nanoemulsions (NEs) for fungal keratitis therapy. The inclusion complexes of MCT oil/alpha-cyclodextrin (alpha CD), AmB/gamma cyclodextrin (gamma CD), and AmB/2-hydroxypropyl-gamma cyclodextrin (HP gamma CD) were characterized using various techniques. The MCT oil/alpha CD complexes were found to be suitable for stabilizing Pickering NEs.

Voriconazole Eye Drops: Enhanced Solubility and Stability through Ternary Voriconazole/Sulfobutyl Ether β-Cyclodextrin/Polyvinyl Alcohol Complexes

Summary: This study aimed to improve the aqueous solubility and chemical stability of voriconazole eye drops by using cyclodextrin and water-soluble polymers. The results showed that sulfobutyl ether beta-cyclodextrin greatly enhanced the solubility of voriconazole, and the addition of polyvinyl alcohol had a synergistic effect on solubilization and stabilization. The developed voriconazole eye drops containing cyclodextrin and polyvinyl alcohol exhibited high mucoadhesion, sustained release, good permeability, and no irritation.

A Current Overview of Cyclodextrin-Based Nanocarriers for Enhanced Antifungal Delivery

Summary: Fungal infections are a serious health issue, especially for patients with weakened immune systems. The low solubility of most antifungal agents can limit their effectiveness. However, their solubility and bioavailability can be improved through complexation with cyclodextrins (CDs). CD-based nanocarriers, such as nanoparticles and liposomes, offer a promising solution for delivering antifungal drugs. These systems have synergistic effects, enhancing drug loading, stability, and permeability through membranes, thereby increasing the bioavailability of antifungals.

Topical dexamethasone delivery to the retina: An aqueous cyclodextrin-based microsuspension

Summary: Topical drug delivery to the eye is challenging, with only a small percentage of the dose reaching the desired target. Invasive drug administration methods are currently used for retinal diseases. This study explores a cyclodextrin-based aqueous eye drop microsuspension as a potential solution, which successfully overcomes barriers to drug permeation and delivers therapeutic drug concentrations to the retina. Results show that the efficacy of this eye drop microsuspension is comparable to that of a intravitreal insert.

Anomalous Properties of Cyclodextrins and Their Complexes in Aqueous Solutions

Summary: Cyclodextrins (CDs) are widely used in pharmaceutical products as solubilizing complexing agents and organic cosolvents. They can self-assemble to form nanoparticles in aqueous solutions, which may cause misinterpretations of experimental results. CDs can enhance drug permeation through membranes without affecting the membrane barrier. This review discusses some unexpected findings encountered during studies of CDs as pharmaceutical excipients.

Novel 3D integrated microfluidic device: Design, construction, and application to the preparation of liposomes for vaccine delivery

This study presents a novel three-dimensional integrated microfluidic device for continuous liposome preparation and purification, which shows potential for industrial production of nano-pharmaceutical applications.
