Summary: Hereditary Breast and Ovarian Cancer syndrome (HBOC) is a genetic condition that predisposes adults to earlier onset of cancer. BRCA1 and BRCA2 mutations are the most common causes of HBOC, but mutations in other genes are also present. A retrospective analysis of 4630 French HBOC suspected cases found pathogenic variants in several genes, including BRCA1, BRCA2, PALB2, RAD51C, TP53, and RAD51D. The study also highlighted the importance of retesting BRCA-negative individuals with an expanded panel and the relevance of functional splicing tests in variant classification.

Summary: This study explored the pathologic lesions associated with architectural distortion (AD) on mammography and found that approximately one-third of lesions associated with AD were invasive, low-grade, ER-positive, HER2-negative, and low stage carcinomas. There was a difference in the incidence of carcinomas between single-feature AD and multi-feature AD, with fewer carcinomas found on core needle biopsy (CNB) for single-feature AD. The rate of malignancy found on excision for benign lesions with a correlate for AD was low. The challenge of radiologic/pathologic correlation and decision-making for excision remains, particularly for patients with multi-feature AD who may have undetected invasive carcinomas.

Summary: Abnormal gene expression plays a key role in cancer development. We hypothesized that in cancer, core promoter sequences could be mutated to interfere with transcriptional factors' interaction, resulting in altered transcriptional initiation, abnormal gene expression, and cancer development. Using triple-negative breast cancer (TNBC) as a model, we identified somatic and germline variants in core promoters of genes that affect gene expression and validated the findings. Comparison with core promoter variation data from unclassified breast cancer revealed TNBC-specific core promoter variants. This study demonstrates the mutability of core promoters in cancer and their impact on transcriptional initiation in TNBC and other cancer types.

Summary: This study aimed to identify cancer-derived RNA biomarkers in plasma extracellular vesicles (EVs) from breast cancer patients. The results showed increased fractions of miRNAs, snRNAs, snoRNAs, and tRFs, but decreased fraction of lncRNAs in breast cancer EVs. Additionally, biomarker models were created that could accurately detect breast cancer and determine tumor estrogen receptor and HER2 status.

Summary: The study analyzed the importance of measuring the size of invasive mammary carcinoma on biopsy and found that while most tumors are larger on excision, 18% are larger or equal on biopsy. Factors predictive of biopsy > excision tumors include stage 1 tumors, especially less than 5 mm, and sampled by mammotome. The study recommends measuring invasive carcinoma on both biopsy and excision.
