Summary: This study found that in IDH-mutated LGGs, most TAMs expressed M1 activation markers CD86 and TNF, with some TAMs co-expressing both M1 and M2-related markers. Higher levels of TAMs expressing M2-related markers were observed in 1p/19q non-codeletion IDH-mutated LGGs compared to 1p/19q codeletion LGGs. Inhibition of M-CSF and TGF beta 1 signaling decreased tumor growth and modulated TAM phenotype in a glioma mouse model, indicating a potential role of these proteins in regulating TAM phenotype in glioma.

The decreased expression level of CCAAT enhancer-binding protein alpha in samples with 1p/19q codeletion led to the downregulation of immune checkpoints Galectin-9 and TIM-3. The loss of chromosome 19q was identified as the main reason for promoting the antitumor immune response in gliomas.

Summary: In this study, the transcriptomic and proteomic profiles of low-grade gliomas (LGG) were investigated using RNA-seq data and tandem mass-spectrometry analysis. The results showed that gene expression in LGG subtypes was mainly influenced by IDH mutations and 1p19q co-deletion. Proteomic analysis identified subtype-specific proteins that could be used to classify the LGG subtypes.

Summary: By generating protein diversity, alternative splicing plays an important role in oncogenesis. The IDH1 and IDH2 mutations, as well as the 1p/19q co-deletion, have become crucial for the molecular classification of diffuse gliomas, which also considers DNA methylation profiling. This study demonstrates the impact of IDH mutation, 1p/19q co-deletion, and glioma CpG island methylator phenotype (G-CIMP) on alternative splicing in diffuse gliomas from The Cancer Genome Atlas (TCGA). The findings suggest that alternative splicing contributes to epigenetic regulation in diffuse gliomas and targeting the genes and pathways affected by alternative splicing may offer potential therapeutic opportunities against gliomas.

Glioma stem cells (GSCs) in conjunction with M2 macrophages are responsible for resistance to and recurrence after alkylating agent chemotherapy in 1p/19q-codeleted gliomas.

Summary: This study developed multiple histogram-based CBF indices and evaluated their association with histopathologic grade in de novo brain tumor patients. The study also investigated the associations between these CBF indices and molecular markers, such as IDH1 mutation, ATRX loss, and 1p/19q co-deletion. The results showed that ASL MRI could provide useful noninvasive imaging assessment of brain tumor grade and molecular marker status, with specific correlations found between certain CBF indices and tumor grade, as well as molecular markers like 1p/19q co-deletion and ATRX retention.

Summary: Angioimmunoblastic T cell lymphoma (AITL) is a common peripheral T-cell lymphoma with poor prognosis, often associated with Epstein-Barr virus (EBV) presence in adjacent B cells. Transcriptome study in AITLs revealed high expression of Bam-HI A rightward transcripts (BARTs) and latency IIc with BNLF2a and BCRF1 gene expression, suggesting EBV's pathogenic role in this pathology.
