Clinical Accuracy of a PLEX-ID Flu Device for Simultaneous Detection and Identification of Influenza Viruses A and B

Respiratory tract infections caused by influenza A and B viruses often present nonspecifically, and a rapid, high-throughput laboratory technique that can identify influenza viruses is clinically and epidemiologically desirable. The PLEX-ID Flu assay (Abbott Molecular Inc., Des Plaines, IL) incorporates multilocus PCR and electrospray ionization-mass spectrometry to detect and differentiate influenza A 2009 H1N1 (H1N1-p), seasonal H1N1 (H1N1-s), influenza A H3N2, and influenza B viruses in nasopharyngeal swab (NPS) specimens. The clinical performance characteristics of the PLEX-ID Flu assay in symptomatic patients were determined in this multicenter trial. A total of 2,617 prospectively and retrospectively collected NPS specimens from patients with influenza-like illness between February 2008 and 28 May 2010 were eligible for inclusion in the study. Each specimen was tested in parallel by the PLEX-ID Flu assay and by the Prodesse ProFLU+ assay (Prodesse Inc., Madison, WI), to detect influenza A and B viruses. Specimens testing positive for influenza A virus by ProFLU+ were subtyped as H1N1-p, H1N1-s, or H3N2 by using the ProFAST+ assay (Gen-Probe Prodesse Inc.). The reproducibility of the PLEX-ID Flu assay ranged from 98.3 to 100.0%, as determined by testing a nine-specimen panel at three clinical sites on each of 5 days. Positive percent agreements (PPAs) and negative percent agreements (NPAs) of the PLEX-ID Flu assay were 94.5% and 99.0% for influenza A virus and 96.0% and 99.9% for influenza B virus, respectively. For the influenza A virus subtyping characterization, the PLEX-ID Flu assay had PPAs and NPAs of 98.3% and 97.5% for H1N1-p, 88.6% and 100.0% for H1N1-s, and 98.0% and 99.9% for H3N2, respectively. The overall agreements between the PLEX-ID and Prodesse ProFLU+/ProFAST+ assays were 97.1 to 100.0%. Bidirectional Sanger sequencing analysis revealed that 87.5% of 96 discrepant results between the PLEX-ID Flu and ProFLU+/ProFAST+ assays were found upon influenza A virus detection and H1N1-p subtyping. The PLEX-ID Flu assay demonstrated a high level of accuracy for the simultaneous detection and identification of influenza A and B viruses in patient specimens, providing a new laboratory tool for the rapid diagnosis and management of influenza A and B virus infections.

Construction of benzyl isothiocyanate-loaded fish skin gelatin-luteolin compound emulsion delivery system, and its digestion and absorption characteristics

Summary: In this study, fish skin gelatin (FSG) and luteolin (LUT) were used as composite emulsifiers, and benzyl isothiocyanate (BITC) was used as a model nutrient for delivery. The FSG-LUT emulsion showed higher stability and embedding rate compared to the FSG emulsion. It also demonstrated resistance to oral and gastric digestion, resulting in higher retention and bioaccessibility of BITC. Furthermore, the ex vivo study in rat intestine showed that the FSG-LUT emulsion facilitated controlled release and absorption of BITC in a specific position.

Comparison of the microbial communities in pits with different sealing methods for Chinese strong-flavor liquor production

Paleogeographic Reconstruction of Precambrian Terranes Reworked by Phanerozoic Orogens: An Example Based on Detrital Zircon REE From Lhasa Terrane in Southern Tibet

Global and regional detrital zircon rare earth element (REE) databases show trends in LREE/HREE and Eu/Eu* that record the crustal evolution of the source and provide a new approach for paleogeographic reconstructions. By analyzing sedimentary and igneous rocks in the Lhasa terrane and detrital zircon samples from the northern margin of Gondwana, it is demonstrated that the Lhasa terrane had an African affinity in the Rodinia-Gondwana supercontinent cycles (ca. 1.4-0.4 Ga).

Glycation of whey protein isolate and stachyose modulates their in vitro digestibility: Promising prebiotics as functional ingredients

Summary: Glycated WPI-stachyose conjugates were prepared and showed increased inhibition of α-amylase activity and delayed glucose dialysis. These conjugates also exhibited reduced digestion by pepsin and trypsin compared to unconjugated WPI, and a 70% lower hydrolysis rate of stachyose. These findings suggest the potential use of glycated conjugates as functional ingredients with sustained-release effect in the gastrointestinal tract.

Improved biogas production from tobacco processing waste via biochar-assisted thermophilic anaerobic digestion

Summary: Tobacco processing waste can be used as feedstock for biogas production through anaerobic digestion. This study examined the thermophilic anaerobic digestion of flue-cured tobacco leaf waste and evaluated the effects of biochar on biogas production. The addition of biochar enhanced methane production by 1.12-1.20 fold at different organic loads. The presence of biochar shortened the lag period and increased the methane production rate. Specific bacterial and methanogenic genera played a crucial role in the digestion process. These findings provide valuable insights for the treatment of tobacco waste.

Collaborative optimization of cutterhead for mass, mode frequency and fatigue life

Summary: Various design methods were adopted to reduce energy consumption and improve service stability and security of the cutterhead, including lightweight design, modal optimization, and life prediction. A multi-disciplinary design optimization methodology was used to determine the design parameters for each discipline, and a Kriging model was employed to approximate the relationship between mode frequency, fatigue life, and design parameters. A collaborative optimization approach was utilized to establish system-level and subsystem-level optimization models, and a genetic algorithm was used for iterative optimization to achieve overall improvement.

Multicenter Study of Colistin Heteroresistance in Carbapenem-Resistant Klebsiella pneumoniae Strains in China
