Summary: The study developed and characterized liposome-based nanoparticles with carvacrol, cinnamaldehyde, citral, and thymol for the therapy of invasive candidiasis. Among the formulations tested, nanoparticles containing cinnamaldehyde, citral, and thymol showed the best characteristics, with high encapsulation efficiencies of citral and carvacrol liposomes. Carvacrol and thymol in liposome-based nanoparticles were non-toxic and maintained their antifungal activity, synergistically working with macrophages to reduce yeast survival.

In Vitro Synergistic Interactions of Isavuconazole and Echinocandins against Candida auris

Summary: Combining isavuconazole with echinocandins showed an overall synergistic interaction against Candida auris, proving to be more effective than monotherapy regimens. Synergism and fungistatic activity were achieved with combinations that included low concentrations of isavuconazole and high concentrations of echinocandins. Results indicated that increasing drug concentrations did not improve antifungal activity once synergy was achieved.

Postantifungal Effect of Antifungal Drugs against Candida: What Do We Know and How Can We Apply This Knowledge in the Clinical Setting?

Summary: This article reviews the pharmacological properties of antifungal agents and focuses on the postantifungal effect (PAFE) of the drugs used to treat Candida infections. The study reveals that the magnitude of PAFE varies depending on the fungal species and the class of antifungal agent. The article compiles the literature information regarding the PAFE of polyenes, azoles, and echinocandins against the Candida species of medical interest, discussing the mechanisms, study methods, and clinical applicability. Furthermore, factors influencing the variability in PAFE are described, with a scarcity of PAFE studies in animal models compared to in vitro studies.

Antimicrobial Peptides with Anti-Candida Activity

Summary: Mycoses are responsible for millions of infections worldwide each year, with invasive candidiasis being the most common and having high morbidity and mortality. The incidence of other Candida species, such as Candida parapsilosis, Candida glabrata, and Candida auris, is increasing. These pathogens often show reduced susceptibility to commonly used antifungal drugs, and the emergence of multi-drug-resistant strains continues to rise. Therefore, the search for new molecules that target these pathogens in a different way is urgent. Antimicrobial peptides, found in nature, have advantages over traditional antifungal agents and are considered a promising option for treating a wide range of microbial infections.

Assessing pH-dependent activities of virulence factors secreted by Candida albicans

Summary: This study aimed to investigate the impact of external pH, substrate nature, and strain origin on the production of protease, lipase, and hemolysin in Candida albicans. The pH value had a significant effect on the detection of protease and lipase, while the activity of hemolysin was not affected by pH. Additionally, the anatomical origin of the isolates was found to be associated with the detected activities.

PK/PD modeling and simulation of the in vitro activity of the combinations of isavuconazole with echinocandins against Candida auris

Summary: In vitro combination of echinocandins and isavuconazole against Candida auris is synergistic, but its clinical efficacy has not been evaluated. This study used a semimechanistic model to characterize the growth and killing of C. auris in response to the combination of these drugs and performed simulations to predict the in vivo response. The model-based simulations suggest that certain combinations may be effective against C. auris, while others may lead to treatment failure.

Virulence of Candida auris from different clinical origins in Caenorhabditis elegans and Galleria mellonella host models

The study found that the aggregating phenotype isolate was less virulent but had higher biofilm-forming capacity compared to non-aggregating isolates. Blood isolates were more virulent in the G. mellonella model, indicating potential heterogeneity in virulence influenced by aggregation phenotype and clinical origin.
