Summary: Lonapegsomatropin, a long-acting GH therapy, has been approved for the treatment of pediatric growth hormone deficiency. It has a longer half-life and requires weekly administration. Clinical trials have shown its positive efficacy in children and adults with GHD, with minimal side effects. The introduction of this long-acting growth hormone therapy may potentially improve patient adherence, quality of life, and clinical outcomes.

Summary: This study investigated the pharmacokinetics and pharmacodynamics of Y-shape branched PEGylated recombinant human growth hormone (YPEG-rhGH) in children with growth hormone deficiency (GHD). The results suggest that YPEG-rhGH is suitable for once-weekly treatment, and a dose of 120-140 mu g/kg/week provides similar efficacy and safety compared to daily rhGH 35 mu g/kg/day.

Summary: This real-world pilot study evaluated the metabolic and endocrinological effects of switching from daily growth hormone (GH) replacement therapy to weekly GH replacement therapy in patients with adult growth hormone deficiency (AGHD). The results suggest that weekly GH replacement therapy may have more benefits compared to daily therapy.

Summary: This study aimed to assess the efficacy and safety of somapacitan in treating growth hormone deficiency (GHD) in children, as well as the burden of treatment. The results showed that weekly injections of somapacitan could reduce the burden for patients and their parents/guardians compared to daily GH injections.

Summary: The objective of this study was to compare the efficacy and safety of once-weekly somatrogon with once-daily Genotropin in Japanese children with growth hormone deficiency (GHD). The study showed that once-weekly somatrogon was comparable to once-daily Genotropin in terms of annualized height velocity (HV) at 12 months. Both treatments had similar safety and tolerability profiles.

Summary: This study compared the efficacy and safety of once-weekly somatrogon with once-daily somatropin in prepubertal children with growth hormone deficiency. The results showed that once-weekly somatrogon was noninferior to once-daily somatropin in terms of height velocity and change in height standard deviation score. Both treatments were well tolerated.

Summary: Linear models were developed to predict mean and peak IGF-I levels based on samples taken on different days after somapacitan administration, showing strong linear relationships between IGF-I sampled on any day and the weekly mean and peak levels. Monitoring IGF-I levels on day 4 and day 2 after dosing provided the most accurate estimates of mean and peak levels, respectively, offering a simple and reliable tool for optimal monitoring of IGF-I.

Summary: This study provides clinical guidance on dosing and titration of somapacitan in patients with adult growth hormone deficiency (AGHD) based on analyses of somapacitan dose-insulin-like growth factor I (IGF-I) responses. It supports higher starting doses for younger patients and women on oral estrogen therapy, and provides information on maintenance dose ratios and appropriate times for IGF-I sampling.

Summary: This study reports a method of genetically fusing recombinant human growth hormone (rhGH) to a thermosensitive elastin-like polypeptide (ELP), resulting in the formation of rhGH-ELP fusion protein. The fusion protein exhibits enhanced stability and sustained release, allowing for the promotion of linear growth and development in rats. This approach has the potential to reduce the frequency of rhGH administration and improve patient convenience.

Summary: Treatment of pediatric growth hormone deficiency with once-weekly somatrogon or once-daily somatropin for 12 months resulted in comparable improvements in health-related quality of life. Results suggest that evaluation of HRQoL could help support treatment decisions in children with pGHD treated with growth hormone.
