Summary: Phosphodiesterase-5 inhibitors (PDE5i) are being studied for their potential in preventing colon cancer. However, their side-effects and interactions with other drugs have been a concern. By modifying the prototypical PDE5i sildenafil, we developed a new analog called malonyl-sildenafil which exhibited reduced lipophilicity and showed promising effects on colon epithelium. This analog suppressed proliferation in the colon epithelium, providing a novel approach to developing a first-in-class drug for colon cancer chemoprevention.

Summary: This study found that Brucella outer membrane protein Omp25 could suppress the production of IFN-beta and its downstream IFN-stimulated genes induced by various DNA viruses or IFN-stimulatory DNA. Omp25 promoted the degradation of cyclic GMP-AMP synthase (cGAS) via the ubiquitin-proteasome-dependent pathway, leading to the inhibition of IFN-beta induction in various mammalian monocyte/macrophages.

Summary: By studying the molecular signaling and positive cardiac contractility induced by beta-blockers in the heart, it was found that carvedilol can promote cardiac inotropy and have a protective effect in diabetic mice. The observations suggest a novel beta(1)AR-NOS3 signaling pathway that may be targeted in therapy for heart diseases with reduced ejection fraction.

Summary: The purpose of this study was to investigate whether raised levels of retinal cyclic guanosine monophosphate (cGMP) in PDE6A(-/-) dogs was reflected in plasma levels. The results showed that cGMP levels were significantly elevated in retinal samples of 2-month-old PDE6A(-/-) dogs, but there was no significant difference in plasma cGMP levels. Therefore, plasma cGMP levels may not be a useful biomarker for the disease at the 2-month timepoint.

Summary: This study investigates the effect of different amounts of metakaolin and zeolite pozzolans on the mechanical properties and durability of concrete containing polypropylene fibers. The results indicate that polypropylene fibers were more effective in enhancing mechanical properties in the zeolite mixtures. The AFM analysis, UPV, density, and permeable voids test results revealed that FRC mixtures incorporating natural zeolite had the most dense microstructure and the least porosity and defects among all examined mixtures.
