Summary: The development of a selective short linear peptide ligand of the IgG Fc region, capable of capturing all four IgG subclasses, was reported. This ligand was discovered through in vitro directed evolution and displayed high affinity for IgG, comparable to commercially employed protein A-based resin, after characterization and validation.

Summary: A novel multicomponent approach combining the isonitrile-tetrazine (4+1) cycloaddition and the Ugi four-component reaction was developed to generate pyrazole amide derivatives. The scope of the reaction and the mechanistic insights of the 4H-pyrazol-4-imine tautomerization were provided. This multicomponent process expands the chemical space of pyrazole amide derivatives and offers a tool for peptide modification and stapling.

Summary: By modifying hyaluronan (HA) through the Ugi reaction, 36 HA-like polysaccharides were generated, and two lead compounds with enhanced CD44 binding were identified. This study confirms the feasibility of HA modification through the Ugi reaction and suggests it as a potential strategy for developing novel binders towards HA receptor such as CD44.

Summary: In this study, pectin was used as a green support to construct an efficient heterogeneous catalytic system based on metal phthalocyanines. The catalytic activity of the copper phthalocyanine-conjugated pectin in the CO2 fixation reaction was investigated. Pectin successfully improved the catalytic properties of the Cu-PcTA.

Summary: This study demonstrates a new method for the synthesis of lipophilic synthetic polyamines (LPAs) and evaluates their anticancer activity in different cancer cell lines. The results show that LPAs with piperazine fragments exhibit comparable anticancer activity to aliphatic LPAs, and the presence of a diglyceride fragment in LPAs appears to be a key factor for their high anticancer activity.

Summary: Virtual screening is an important method in drug discovery that relies on a virtual library of compounds. Ugi reaction is a reliable multi-component reaction that produces a peptidomimetic scaffold, and recent research has renewed interest in it due to its potential for further chemical modifications. A library of small molecules derived from Ugi reaction was collated for virtual screening, and the analysis shows that these molecules are 'clean', drug-like, and have a structurally distinct space compared to FDA-approved oral drugs and inhibitors of protein-protein interactions. The molecules in this library consist of diverse ring systems, many of which have not been exploited for drug design, making it a valuable resource for virtual screening.

Summary: The Ugi four-component reaction (Ugi-4CR) is a highly influential multicomponent reaction that has been widely used in the synthesis of diverse heterocycle molecules. It involves the use of amine, aldehyde or ketone, isocyanide, and carboxylic acid to produce α-acetamido carboxamide derivatives, significantly advancing the field of isocyanide-based multicomponent reactions. Alternative reagents for the components have been explored, making the Ugi reaction a versatile method for the synthesis of biologically active molecules and complex scaffolds. This review focuses on the use of alternative components in Ugi multicomponent reactions, particularly over the past decade.

Summary: A Biginelli, hydrolysis and Ugi four-component reaction sequence was developed to synthesize novel dihydropyrimidinone (DHPM) bisamides. An unprecedented but moderate 1,6 asymmetric induction was observed for this compound class. A detailed study was carried out to investigate the effects of various substitutions on the diastereoselectivity. The results showed that steric congestion caused sluggish Mumm rearrangement, leading to the formation of side products by trapping the imidate intermediate with methanol.
