Analysis of sibutramine metabolites as N-trifluoroacetamide and O-trimethylsilyl derivatives by gas chromatography-mass spectrometry in urine

A method for identifying the metabolites of sibutramine 1-(4(chlorophenyl)-N,N-dimethyl-alpha-(2-methylpropyl))cyclobutanemethanamine) in urine, utilizing a double derivatization strategy, with N-methyl-N-(trimethylsilyl)-trifluoroacetamide and N-methyl-bis-(trifluoroacetamide), in gas chromatography/mass spectrometry is proposed. This methodology results in mass spectra with at least three fragments in abundance superior to 20%, attending the World Anti-Doping Agency identification criteria for qualitative assays. The characterization of the derivatives was obtained through two ionization modes: Chemical Ionization and Electron Impact ionization, both in full scan mode. Sibutramine was administered to 5 (five) volunteers and the excretion profile followed for 92 h. Routine analytical, hydroxy-cyclobutane-bis-nor-sibutramine which becomes the more abundant metabolite in the first 10 h and hydroxy-isopropyl-bis-nor-sibutramine which becomes the most abundant after 40 h, were proposed for doping monitoring. Summary: The study aimed to identify in vivo phase I metabolites of five psychoactive substances using a zebrafish water tank experimental setup and high-resolution mass spectrometry. Results showed that alpha-PVP, N-ethylpentylone, ethylone, and 4-CDC can be transformed into phase I metabolites, while methylone appears to be a potent CYP450 inhibitor in zebrafish. The approach was found to be simple, cost-effective, and efficient for studying the metabolism of synthetic cathinones.

Effective treatment and prevention of attempted suicide, anxiety, and aggressiveness with fluoxetine, despite proven use of androgenic anabolic steroids

Summary: This study details the treatment of a man who attempted suicide after experiencing symptoms of anxiety and aggressiveness associated with the use of AAS. Regular use of fluoxetine was effective in preventing anxiety, depression, aggressiveness, and suicide ideation, even in the presence of AAS. Urinary concentration of androgens, AAS metabolites, and fluoxetine were monitored through urine samples analysis at the Brazilian Laboratory of Doping Control.

Prior to 2014, LSD was the main substance found in seized blotter papers, while an increase in NBOMe compounds was observed from 2014 to 2017, with NBOH compounds emerging in coastal regions from 2016.

Summary: The use of androgenic-anabolic steroids (AAS) can lead to psychiatric symptoms such as insomnia, anxiety, and increased aggressiveness. This study on bodybuilders found that the majority of participants experienced symptoms of agitation, insomnia, increased aggressiveness, or depression. There was no significant difference in psychiatric symptoms between males and females, and no direct association between symptoms and drug dosage.

Hansen Solubility Parameters Applied to the Extraction of Phytochemicals

Summary: In many analytical chemical procedures, organic solvents are used to separate, extract, or isolate target phytochemical analytes. The selection of extraction solvents is based on solubility differences, cost, and safety. Hansen Solubility Parameters are used to predict the miscibility of compounds, saving time and costs in method development. This review discusses how Hansen Solubility Parameters have been incorporated into classical and emerging extraction techniques, saving resources and avoiding unnecessary solvent wasting.
