Summary: We aimed to establish an LC-MS/MS method for quantifying bevacizumab content in biological fluids using a surrogate peptide generated through trypsin hydrolysis and an isotopically labeled peptide as an internal standard. The method involved a series of pre-treatment processes and had a total duration of approximately 2.5-3 h. The validated method showed a linear range of 0.2 μg/mL to 300 μg/mL in different matrices, with satisfactory precision and accuracy.

Summary: Apolipoprotein A-I mimetic peptide L37pA modulates the gene expression program of interferon alpha, reducing inflammatory pathways activation and inducing an anti-inflammatory effect in the liver. This modulation allows for the silencing of interferon alpha expression mediated by an adeno-associated virus, preventing lethal hematological toxicity in mice. The proteomic analysis confirms the impact of L37pA on several inflammatory pathways and indicates the activation of LXR/RXR and PPPAR alpha/gamma nuclear receptors.

Summary: The increasing awareness of consumers on sustainable food chains has caused a shift in consumption from animal protein sources to plant protein sources. Soy, which is both relevant for human food use and animal feed, has a high protein content but is accompanied by antinutritional factors such as Kunitz's trypsin inhibitor (KTI). A direct label-free liquid chromatography-mass spectrometry (LC-MS) method has been developed to identify and quantify Kunitz inhibitor KTI3 in soybean and its derivative products, using a marker peptide specific to the protein of interest. The results of the LC-MS method were compared with spectrophotometric trypsin inhibition, demonstrating the complementarity of these two types of analysis.

Summary: This study investigated the influence of different proteoforms of vitamin D-binding protein (VDBP) on quantification using liquid chromatography-tandem mass spectrometry (LC-MS/MS). The results showed that VDBP proteoforms do not cause bias and a family of proteins can be accurately quantified using shared peptides. Assigning a reference value in a standard reference material can improve the accuracy of VDBP quantification in research and clinical samples.

Summary: This study identified and investigated the effects of a novel multifunctional peptide from Lingzhi on melanogenic enzymes in melanoma cells through a targeted-proteomics approach. The synthesized octapeptide exhibited strong antioxidant activity and regulated the expression of specific proteins involved in pigmentation pathways. These findings contribute to the understanding of the molecular functions of the multifunctional octapeptide on melanogenic enzymes and support its potential as a therapeutic and cosmetic ingredient.
