Summary: The purpose of this study was to investigate the effect of CYP3A4 variants on tofacitinib metabolism and the interaction of tofacitinib with resveratrol. It was found that the clearance rate of CYP3A4.18 variant was significantly decreased compared with CYP3A4.1, and the CYP3A4.28 variant was changed, but not statistically significant. Resveratrol was found to have an IC50 value of 15.67 mu M with a non-competitive mechanism in rat liver microsomes and 8.88 mu M in human liver microsomes. In vivo study showed that the AUC(0-infinity) and Cmax of tofacitinib increased significantly in the presence of resveratrol. In conclusion, resveratrol has a significant impact on the metabolism of tofacitinib.

Summary: This study aimed to evaluate the absolute bioavailability of cyclosporine in cats by comparing the pharmacokinetic profiles of oral and intravenous administration. The results showed that the medium dose of cyclosporine had great potential to be absorbed in the body. Monitoring the concentration of cyclosporine is necessary to prevent side effects in clinical use. Taking a blood sample 4 hours after oral administration is recommended for better estimation of absorption extent and prevention of side effects. Rating: 7/10.

Summary: The pharmacokinetics of nalbuphine were characterized in patients with varying degrees of liver dysfunction. The results showed that in patients with moderate/severe liver dysfunction, the elimination half-life and volume of distribution of nalbuphine were significantly increased. Multivariate analysis also revealed associations between weight, platelet count, total bilirubin, and the clearance and half-life of nalbuphine.

Summary: A study was conducted to investigate the pharmacokinetic compatibility of two components in the traditional Chinese medicine prescription Jinlingzi San. Results showed a decrease in elimination of one limonoid and alkaloids in the Jinlingzi San group compared to the single herb group, with an increase in exposure of the alkaloids to some extent. The study demonstrated a synergistic effect of combining Fructus Toosendan with Rhizoma Corydalis on the pharmacokinetics of seven bioactive components, providing new insights into the compatibility mechanism of Jinlingzi San.

Summary: This study aimed to investigate the phytochemical profile of alkaloids in N. officinalis total alkaloids and determine their pharmacokinetic behaviors. The results showed that 55 alkaloids were identified in N. officinalis total alkaloids, with 25 alkaloids absorbed by rats after oral administration. The established LC-ESI-MS/MS method was successfully applied for the simultaneous determination of major active alkaloids in rat plasma, providing valuable insights for pre-clinical and clinical investigations.

Summary: An efficient synthesis of a tricyclic analog of artemisinin was achieved, with one enantiomer exhibiting antimalarial activity. Unexpectedly, the C-5 hydroxyl group was unreactive, and this unusual reactivity was attributed to a close intramolecular hydrogen bond revealed by X-ray crystallography. Several newly synthesized compounds showed moderate antimalarial activity in vitro.

A machine learning model was used to estimate the experimental Delta T-m value.
