Separation of mesenchymal stem cells with magnetic nanosorbents carrying CD105 and CD73 antibodies in flow-through and batch systems

The aim of this study is to develop magnetically loaded nanosorbents carrying specific monoclonal antibodies (namely CD105 and CD73) for separation of mesenchymal stem cells from cell suspensions. Super-paramagnetic magnetite (Fe3O4) nanoparticles were produced and then coated with a polymer layer containing carboxylic acid functional groups (average diameter: 153 nm and polydispersity index: 0.229). In order to obtain the nanosorbents, the monoclonal antibodies were immobilized via these functional groups with quite high coupling efficiencies up to 80%. These nanosorbents and also a commercially available one (i.e., microbeads carrying CD105 antibodies from Miltenyi Biotec., Germany) were used for separation of CD105(+) and CD73(+) mesenchymal stem cells from model cell suspension composed of peripheral blood (97.6%), human bone marrow cells (1.2%) and fibroblastic cells (1.2%). The initial concentrations of the CD105(+) and CD73(+) cells in this suspension were measured as 5.86% and 6.56%, respectively. A flow-through separation system and a very simple homemade batch separator unit were used. We were able to increase the concentration of CD105(+) cells up to about 86% in the flow-through separation system with the nanosorbents produced in this study, which was even significantly better than the commercial one. The separation efficiencies were also very high, especially for the CD73(+) cells (reached to about 64%) with the very simple and inexpensive homemade batch unit. (c) 2007 Elsevier B.V. These compounds exerted anticancer effects through different cell death mechanisms.

Summary: mTOR pathway is an important kinase that regulates cell cycle, protein synthesis, and energy metabolism. In this study, rapamycin treatment effectively inhibited mTOR and P70S6K activity in TCam-2 cells, leading to suppressed cell proliferation and induced apoptosis. These findings contribute to the development of new treatment approaches for seminoma and preservation of male fertility.

Secondary metabolites from the underground parts of Valeriana sisymbriifolia Vahl. and their in vitro cytotoxic activities

Summary: Cytotoxic activity-guided isolation studies identified 12 secondary metabolites from Valeriana sisymbriifolia, including two undescribed iridoids and two unreported sesquiterpene lactones. Extensive analysis was performed to establish the chemical structures of the isolates. In vitro cytotoxicity evaluations showed that certain compounds exhibited promising activity against cancer cell lines, with valtrate and 7-homovaltrate being particularly potent against multiple cell lines.

Effects of SPARC and Possible Receptors on Colon Cancer Cell Line

Summary: The study aimed to observe the effects of exogenous SPARC on colon cancer cell line HT-29, investigate the possible receptors stabilin-1 and integrin alpha v beta 3, and determine the receptor numbers. The appropriate doses of SPARC and its inhibitor cilengitide were determined, caspase 3 expressions were measured, and gene expression levels and receptor numbers were detected. The results showed that exogenous SPARC reduced proliferation and induced apoptosis in colon cancer cells, with integrin alpha v beta 3 considered as a possible mediating receptor for SPARC. Quantification of surface receptors per cell can be a marker for anticancer treatments.

Summary: This study synthesized and characterized different Pc derivatives for anti-cancer efficacy, showing varied effects on breast cancer and endothelial cell lines. In particular, InPc derivatives exhibited enhanced effects on the mitochondrial network, while non-peripheral substitutions led to decreased anti-cancer activity. Further research is needed to determine ideal Pc candidates for anti-cancer PDT treatments.

Kappa Deleting Recombination Excision Circle (KREC) Immunoglobulin Gene Rearrangement Mechamism and Importance in Transplants

Summary: KREC is a molecular structure that occurs in response to B lymphocytes, protecting the immune system against various antigens and serving as an indicator of early immune response, especially in immunodeficiency patients. It can be measured through molecular analysis to show B cell output from the bone marrow and has been increasingly studied in clinical settings. The mechanism of KREC occurrence, importance of KREC measurement, and its role in disease treatment are the main focus of this review.
