Summary: Analytical expressions for retention time and peak compression factor are deduced by assuming quadratic solvent strength model and multilinear gradient elution. A program for the optimization of multilinear gradient profile is written and applied to search for a gradient profile for the separation of twelve compounds that are degraded from lignin. Results show that a better separation of the compounds is achieved under an S-shaped multilinear gradient profile than that obtained under linear gradient profile.

This study introduces a Tecan Freedom EVO high-throughput purification tool that enables continuous gradient elution capability, leading to improved performance and consistency between RoboColumns and larger column formats.

Summary: Ion-pairing reversed-phase liquid chromatography (IP-RPLC) is the reference separation technique for characterizing oligonucleotides (ONs) and their related impurities. This study aimed to understand the retention mechanism of ONs, evaluate the applicability of the linear solvent strength (LSS) retention model, and explore ultra-short columns for ONs separation. It was found that 5-35 mm column lengths were appropriate for most gradient separation conditions. Baseline separation of three model ONs mixtures was achieved in as little as 30 seconds on the 5 mm column. This proof-of-concept work paves the way for future investigations using more complex therapeutic ONs and their related impurities.

Summary: Recent advancements in cell engineering and bioreactor engineering have led to increased concentrations of monoclonal antibodies (mAbs) in downstream process (DSP). A method for measuring high protein concentration (C-P) using optical rotation (OR) was developed and successfully applied in monitoring C-P in cell culture solutions. The results showed good agreement with the standard Protein A HPLC method, indicating the potential of OR as an efficient process analytical tool (PAT) for DSP.

Summary: With the increasing demand for therapeutic monoclonal antibodies, in silico downstream process development based on mechanistic modeling of chromatography separation process is becoming popular. However, model calibration, especially obtaining input values for binding kinetic parameters, can be time-consuming. This study proposes an accelerated approach to estimate the input values for binding kinetic parameters using Yamamoto coefficients A and B obtained directly from model calibration linear gradient elution runs. The proposed method is more efficient and accelerated than current approaches, accelerating the development and implementation of mechanistic modeling for process chromatography.

Summary: A continuous downstream process for monoclonal antibodies was developed using periodic-counter current chromatography (PCCC) for capture, a batch reactor for low pH virus inactivation (VI), and flow-through chromatography (FTC) for polish. Different runs with varying feed rates and concentrations were performed, achieving good recovery and purity values. The process was found to be flexible and stable for feed fluctuations, requiring only a few surge or pool tanks in addition to the final product pool tank.

Summary: Biosorption using phosphorylated hazelnut shell waste (FHS) is a promising method for lithium removal and recovery. This study investigated the performance of a continuous-flow packed-bed column system using FHS for ultra-low level lithium removal. The results showed that the lihium biosorption capacity of the FHS column was unaffected by the bed height, but decreased with increasing flow rate.

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.

Summary: A chitosan-alginate sponge with multiple cross-linking networks was developed for adsorption and enrichment of anionic dyes from food samples. The sponge showed better three-dimensional network structure, mechanical behavior, and stability compared to pure chitosan and alginate sponges. The adsorption performance and selectivity of the sponge were systematically studied, and it maintained high adsorption capacity and selectivity for anionic dyes. The sponge also showed satisfactory reusability and was successfully combined with high-performance liquid chromatography for the enrichment and detection of anionic dyes in candy and cocktail samples.

Summary: Analysis of in vivo exposure to traditional Chinese medicine using mass spectrometry is helpful for identifying effective ingredients and developing new drugs. This study focused on Forsythia suspensa (Lian Qiao) and used metabolomics technology to explore its in vivo exposure. By comparing the differences before and after administration, a complete set of nontargeted analysis strategies for TCM exposure was established. The study identified 393 Lian Qiao-related chemical components in mice after administration, including prototypes and metabolites. This study provides a reference for research on the active ingredients of Lian Qiao in vivo.
