Summary: In the respiratory tract, extracellular 3',5'-cAMP is metabolized by enzymes to produce 5'-AMP, adenosine, and inosine, affecting airway smooth muscle contraction and relaxation. This extracellular enzymatic system plays a role in respiratory physiology and pathophysiology by generating interstitial adenosine from 3',5'-cAMP.

Summary: Neuronal activity leads to broad metabolic changes, including increased glucose metabolism, in the dentate granule cell layer. Stimulation triggers the conversion of endogenous inosine into pentose phosphates via the action of purine nucleotide phosphorylase. Mass spectrometry imaging allows for the study of rapid metabolic responses in brain slices.

Summary: The study reveals that neuronal activity induces metabolic changes in neurons, including increased glycolytic activity and conversion of endogenous inosine. The purine nucleotide phosphorylase (PNP) reaction is crucial for the neuronal response to stimulation, and inhibition of PNP leads to energy impairment in neurons during recovery.

Summary: We identified the pathological mechanisms of aberrant metabolism in the cerebral cortex of patients with neuropathic pain using metabolomics and proteomics analyses. Chronic constriction injury resulted in significant dysregulation of metabolites and proteins involved in alanine, aspartate and glutamate metabolism, GABAergic synapse, and retrograde endocannabinoid signaling. Our study provided insights on the roles of the endogenous pain-modulatory system in neuropathic pain.

Summary: We elucidated the aberrant metabolism of the cerebral cortex in neuropathic pain induced by chronic constriction injury (CCI) using metabolomics and proteomics analyses. Integrative analyses showed that the differentially expressed metabolites and proteins were primarily involved in alanine, aspartate and glutamate metabolism, GABAergic synapse, and retrograde endocannabinoid signaling. Our study revealed the alterations of the endocannabinoid system and purinergic system in the CCI group, providing a novel perspective on the roles of the endogenous pain-modulatory system in the pathological mechanisms of neuropathic pain.

Summary: The study showed that synoviocytes of OA and RA patients have the ability to synthesize adenosine/inosine, with adenosine predominantly responsible for the anti-inflammatory effects. Inhibiting CD73 increased IL-6 and decreased IL-10, while inhibiting ADA reduced IL-6 and induced IL-10. A(2A)- and A(2B)AR play a crucial role in mediating anti-inflammatory or pro-inflammatory effects.

Summary: This study presents an injectable hydrogel strategy to reshape the adenosinergic axis by converting adenosine into inosine for potent cancer therapy. The strategy boosts ATP induction and tumoricidal effect while catalyzing adenosine into inosine to regulate accumulation and enhance immunity. By narrowing the adenosine pool, enriching the ATP reservoir, and transforming immunosuppressive adenosine into immunopotentiator inosine, this approach effectively suppresses tumor progression.

Summary: Adenosine-to-inosine (A-to-I) RNA editing is a gene regulatory mechanism that is widespread in many metazoans and plays a key role in diverse biological processes. The ADAR family of enzymes, including ADAR1, ADAR2, and ADAR3, catalyzes this editing process. A-to-I editing can generate new protein isoforms and influence RNA processing pathways. ADAR1 is especially important in preventing aberrant activation of the innate immune system. However, the identification of A-to-I editing sites remains challenging, and nanopore sequencing technology shows promise in addressing this issue.

Summary: This study evaluated the immunization effects of recombinant Schistosoma mansoni enzymes (AK and HGPRT) or a mixture of the two enzymes. The results showed a reduction in eggs in the feces and adult worms in the group immunized with the enzyme mixture. The AK group also showed a decrease in hepatic granulomas and eggs. Therefore, immunization with these enzymes can contribute to schistosomiasis control and reduce the pathology of the disease.

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.
