Selective Densitometric Determination of Four alpha-Aminocephalosporins Using Ninhydrin Reagent

A simple, selective, and precise densitometric method for analysis of four alpha-aminocephalosporins, namely cefaclor monohydrate, cefadroxil monohydrate, cefalexin anhydrous, and cefradine anhydrous, both in bulk drugs and in formulations was developed and validated. The method employed thin-layer chromatography (TLC) aluminium sheets precoated with silica gel G 60 F-254 as the stationary phase. The solvent system consists of ethyl acetate-methanol-water with different ratios for all studied drugs (R-f values of 0.40-0.60). The separated spots were visualized as blue to violet color after spraying with ninhydrin reagent. The linear regression analysis data for the calibration plots of all studied drugs produced a good linear relationship with correlation coefficients ranging from 0.9990 to 0.9996 and coefficients of determination ranging from 0.9986 to 0.9992 over the concentration range 2-10 mu g/spot. The limits of detection and quantitation for all studied drugs ranged from 0.09 to 0.23 and from 0.27 to 0.84 mu g/spot, respectively. The developed method was applied successfully for the determination of the studied drugs in their pharmaceutical dosage forms with good precision and accuracy. Also, the method can be employed as a promising stability-indicating assay.

Summary: Avanafil is a new PDE-5 inhibitor used for treating erectile dysfunction, often coadministered with antihypertensive drugs like Nimodipine. A new electrochemical method using nanoparticle-modified electrodes was developed to simultaneously determine the levels of Avanafil and Nimodipine in human serum samples. The study found that coadministration of Avanafil and Nimodipine may lead to a significant increase in Avanafil serum concentration, highlighting the importance of dose adjustment when these drugs are used together.

Summary: Diabetes is a risk factor for sexual dysfunction in men, specifically increasing the risk of erectile dysfunction. Canagliflozin is a commonly used antidiabetic drug for type-2 diabetes and can be combined with phosphodiesterase 5 inhibitors to alleviate erectile dysfunction. This study developed a high-performance thin-layer chromatographic method to simultaneously determine the concentrations of tadalafil and canagliflozin in human plasma and demonstrated a significant interaction between the two drugs when administered simultaneously.

Anatase titanium oxide nanoparticles and multi-walled carbon nanotubes-modified carbon paste electrode for simultaneous determination of avanafil and doxorubicin in plasma samples

Summary: Patients receiving anticancer drugs may experience loss of sexual desire and erectile dysfunction as adverse effects. It is important to recognize possible pharmacokinetic interactions when administering drugs to cancer patients. A sensitive sensor using carbon paste electrode modified with titanium dioxide nanoparticles and multi-walled carbon nanotubes was designed for the simultaneous determination of the anticancer drug avanafil and doxorubicin in rabbit plasma samples. The modified electrode showed improved sensitivity and separation between the oxidation peaks of AVN and DOX. The method was effective for analyzing AVN in real rabbit plasma samples and monitoring the possible pharmacokinetic interactions between AVN and DOX.
