Ephrin A1 promotes proliferation of bovine endometrial cells with abundant expression of proliferating cell nuclear antigen and cyclin D1 changing the cell population at each stage of the cell cycle

Ephrin A1 has a role in a variety of biological events, including cell proliferation, differentiation, migration, and angiogenesis. Ephrin A1 expression is abundant in trophoblasts and endometrial cells during the implantation period; however, its intracellular activities have not yet been reported in bovine endometrial (BEND) epithelial cells. The aim of this study was to identify the functional role of ephrin A1 in BEND cells, which have served as a good model system for investigating the regulation of signal transduction following treatment with interferon- (IFNT) in vitro. Supplementation of ephrin A1 to BEND cells increased cell proliferation and increased levels of proliferating cell nuclear antigen and cyclin D1 protein in BEND cell nuclei. To investigate intracellular mechanisms regulated by ephrin A1, we performed Western blot analysis focused on mitogen-activated protein kinase (MAPK) and phosphoinositide 3-kinase (PI3K) signaling, which are significantly involved in the successful maintenance of pregnancy. Ephrin A1 dose-dependently increased phosphorylation of extracellular signal-regulated kinases (ERK)1/2, c-Jun N-terminal kinases (JNK), P38, protein kinase B (AKT), P70S6K, S6, and cyclin D1, and the activated proteins were suppressed by pharmacological inhibitors including wortmannin (a PI3K inhibitor), U0126 (an ERK1/2 inhibitor), and SP600125 (a JNK inhibitor). Among ephrin A1 receptors, abundant expression of EPHA2 and EPHA4 messenger RNA was detected in BEND cells by reverse transcription polymerase chain reaction analysis. Furthermore, tunicamycin-induced endoplasmic reticulum (ER) stress was inactivated by ephrin A1 treatment of BEND cells. Our findings suggest that ephrin A1 promotes the development of BEND cells and likely enhances uterine capacity and maintenance of pregnancy by activating MAPK and PI3K signaling cascades and by restoring ER stress.

Embryonic exposure to chloroxylenol induces developmental defects and cardiovascular toxicity via oxidative stress, inflammation, and apoptosis in zebrafish

Summary: Chloroxylenol is a widely used anti-microbial compound, and recent studies have emphasized the need to assess its potential for ecotoxicity due to increased usage during the coronavirus pandemic and ban on other antimicrobial ingredients. Although the detrimental effect of chloroxylenol on zebrafish viability has been reported, the mechanisms underlying its toxicity are not well understood. This study used the zebrafish model to investigate the responses to chloroxylenol and predict its toxicity to human health and ecology. The findings suggest the risks of chloroxylenol exposure and highlight the importance of further ecotoxicological studies.

Developmental defects induced by thiabendazole are mediated via apoptosis, oxidative stress and alteration in PI3K/Akt and MAPK pathways in zebrafish

Summary: This study demonstrated the developmental toxicity of thiabendazole, a benzimidazole fungicide, using zebrafish as a representative toxicological model. It showed various morphological malformations, as well as triggered apoptosis, reactive oxygen species production, and inflammatory response in zebrafish larvae exposed to thiabendazole. The study also revealed the significant changes in PI3K/Akt and MAPK signaling pathways, as well as toxicity in various organs, such as cardiovascular, neurotoxicity, and hepatic and pancreatic toxicity.

Dietary supplementation with L-citrulline improves placental angiogenesis and embryonic survival in gilts

Summary: This study used gilts as an animal model to investigate the effects of dietary supplementation with L-citrulline (Cit) on placental angiogenesis and embryonic survival. Compared to the control group, Cit supplementation significantly increased the number of viable fetuses, placental blood vessels, placental weight, and fluid volumes. Additionally, Cit supplementation enhanced the synthesis of NO and polyamines, as well as the expression of angiogenic factors and aquaporins in the placenta. Overall, dietary Cit supplementation improved conceptus development and survival by enhancing placental NO and polyamine synthesis and promoting angiogenesis.

Folpet promotes apoptosis of bovine mammary epithelial cells via disruption of redox homeostasis and activation of MAPK cascades

Summary: This study aimed to investigate the harmful effects of folpet on bovine mammary glands and milk production using MAC-T cells. The results showed that folpet exhibited cytotoxicity against MAC-T cells, causing apoptosis and cell death. Furthermore, folpet treatment induced oxidative stress and activated MAPK signaling pathways. This is the first report highlighting the detrimental impacts of folpet on bovine mammary glands and the dairy industry, providing insights into the intracellular mechanisms using MAC-T cells.

Oryzalin impairs maternal-fetal interaction during early pregnancy via ROS-mediated P38 MAPK/AKT and OXPHOS downregulation

Summary: Through experiments on porcine uterine epithelial cells and trophectoderm cells, we found that Oryzalin can inhibit cell growth and proliferation, and also affect the cell cycle progression. In addition, it can induce apoptosis and disrupt mitochondrial membrane polarity. We also confirmed that Oryzalin can decrease adenosine triphosphate (ATP) production and increase reactive oxygen species (ROS) generation in cells. In summary, Oryzalin causes implantation failure through ROS-mediated cellular signaling regulation and inhibition of migration potential.

Picolinafen exposure induces ROS accumulation and calcium depletion, leading to apoptosis in porcine embryonic trophectoderm and uterine luminal epithelial cells during the peri-implantation period

Summary: This study found that picolinafen has a significant impact on the viability of porcine trophectoderm and luminal epithelial cells involved in the implantation process during early pregnancy. It increased apoptosis, disrupted mitochondrial function, and caused intracellular ROS accumulation, which led to a reduction in calcium levels. Furthermore, it inhibited the migration of porcine trophectoderm cells. These effects were mediated by the activation of the MAPK and PI3K signaling pathways.

It also affected normal growth by abnormal expression of cell cycle regulatory genes and increased oxidative stress, inflammation, and apoptosis.
