Summary: This study investigated the mechanisms underlying CysLT1R-mediated mast cell activation. The results showed that CysLT1R plays a major role in accelerating mast cell activation. CysLT1R enhances mast cell activation by accelerating IgE-induced signal transduction, enabling the co-regulation of rapid degranulation and delayed synthesis of inflammatory mediators.

Summary: The skin is equipped with immune cells like mast cells and CD8 T cells which play a role in recognizing and responding to environmental challenges. These cells are essential for maintaining tissue balance and regulating the immune response in skin diseases. Studies have shown that mast cells can influence the migration and function of CD8 T cells, while CD8 T cells can also activate mast cells. The close proximity between mast cells and CD8 T cells has been observed in dermatoses such as alopecia areata. This review summarizes the bidirectional interactions between human mast cells and CD8 T cells, focusing on their involvement in psoriasis, atopic dermatitis, and vitiligo, and discusses unresolved questions.

Summary: In this study, the effects of mast cell extracellular traps (MCETs) released from mast cells after infection with periodontal pathogen Fusobacterium nucleatum were investigated. F. nucleatum induced the release of MCETs from mast cells, and these MCETs expressed macrophage migration inhibitory factor (MIF). Importantly, MIF bound to MCETs induced the production of proinflammatory cytokines by monocytic cells. These findings suggest that MIF expressed on MCETs released upon infection with F. nucleatum promotes inflammatory responses associated with the pathogenesis of periodontal disease.

Summary: This study confirms the direct activation of α7nAChRs in macrophages using macrophage-specific α7nAChR-deficient mice. The α7nAChR-specific agonist, GTS-21, protects injured kidneys in wild-type mice but not in macrophage-specific α7nAChR-deficient mice. Single-cell RNA-sequencing of the spleen reveals an increase in macrophage-macrophage interactions induced by α7nAChR activation.

Summary: In this review, the regulatory effects of endocannabinoids on the activation of basophils and mast cells are summarized. The presence of the endocannabinoid system and cannabinoid receptors in these cells, as well as their unique signaling characteristics, are emphasized. The interactions between endocannabinoids and FceRI signaling cascades are discussed in detail. Furthermore, important considerations and future perspectives in the study of the effects of endocannabinoids in basophils and mast cells are presented.
