Spectrum of Neuroradiologic Findings Associated with Monogenic Interferonopathies

P. Benjamin, S. Sudhakar, F. D'Arco, U. Lobel, O. Carney, C-J Roux, N. Boddaert, C. Hemingway, D. Eleftheriou, K. Mankad

Summary: Genetic interferonopathies are a diverse group of disorders caused by dysregulated interferon expression, increasingly recognized in the differential diagnosis of children with recurrent inflammatory phenotypes. Neuroimaging plays a vital role in the diagnosis, and understanding the wide spectrum of neuroimaging features associated with these disorders can aid in earlier diagnosis and treatment strategies.

Terminal 6q deletions cause brain malformations, a phenotype mimicking heterozygous DLL1 pathogenic variants: A multicenter retrospective case series

Marion Lesieur-Sebellin, Marianne Till, Philippe Khau Van Kien, Berenice Herve, Nicolas Bourgon, Celine Dupont, Anne-Claude Tabet, Mathilde Barrois, Aurelie Coussement, Laurence Loeuillet, Eve Mousty, Vuthy Ea, Amal El Assal, Laura Mary, Sylvie Jaillard, Claire Beneteau, Claudine Le Vaillant, Charles Coutton, Francoise Devillard, Carole Goumy, Amelie Delabaere, Sylvia Redon, Yves Laurent, Audrey Lamouroux, Jerome Massardier, Catherine Turleau, Damien Sanlaville, Vincent Cantagrel, Pascale Sonigo, Francois Vialard, Laurent J. Salomon, Valerie Malan

Summary: Terminal 6q deletion is a rare genetic condition associated with neurodevelopmental disorders and intellectual disabilities. This study collected ultrasound findings from 22 fetuses with pure 6qter deletion and compared them with previous cases and patients with DLL1 pathogenic variants. The results showed common brain structural abnormalities in all fetuses, highlighting the importance of prenatal sonographic anomalies and chromosomal microarray analysis in diagnosing this syndrome.

Recessive PRDM13 mutations cause fatal perinatal brainstem dysfunction with cerebellar hypoplasia and disrupt Purkinje cell differentiation

Marion Coolen, Nami Altin, Karthyayani Rajamani, Eva Pereira, Karine Siquier-Pernet, Emilia Puig Lombardi, Nadjeda Moreno, Giulia Barcia, Marianne Yvert, Annie Laquerriere, Aurore Pouliet, Patrick Nitschke, Nathalie Boddaert, Antonio Rausell, Ferechte Razavi, Alexandra Afenjar, Thierry Billette de Villemeur, Almundher Al-Maawali, Khalid Al-Thihli, Julia Baptista, Ana Beleza-Meireles, Catherine Garel, Marine Legendre, Antoinette Gelot, Lydie Burglen, Sebastien Moutton, Vincent Cantagrel

Summary: Pontocerebellar hypoplasias (PCHs) are congenital disorders characterized by cerebellar and brainstem abnormalities. Recent research identified bi-allelic mutations in PRDM13 as causing olivopontocerebellar hypoplasia syndrome, suggesting transcriptional dysregulation in neuronal fate specification may contribute to the condition.

Slavotinek, Christian Beetz, Dan Diego-Alvarez, Kapil Kampe, Nicole Fleischer, Zain Awamleh, Rosanna Weksberg, Robert Kopajtich, Thomas Meitinger, Jehan Suleiman, Ayman W. El-Hattab

Summary: This study further investigates Suleiman-El-Hattab syndrome, revealing its characteristics and potential mechanisms. The results show that TASP1 deficiency can lead to dysregulation of HOX gene expression and the TFIIA complex, and the methylation pattern suggests that Suleiman-El-Hattab syndrome can be categorized as a type of histone modification disorder.

S. Neumane, A. Lesage, V. Dangouloff-Ros, R. Levy, C. -J. Roux, M. P. Robert, D. Bremond-Gignac, N. Boddaert

Summary: The study retrospectively reviewed the MR imaging of 27 children to describe the signal of common pediatric orbital lesions on arterial spin-labeling and evaluate its ability to discriminate malignant from benign masses. The arterial spin-labeling perfusion patterns were classified into homogeneous hypoperfusion, heterogeneous hyperperfusion, and homogeneous intense hyperperfusion. Arterial spin-labeling was found to be valuable in improving the diagnostic confidence of some orbital lesions.
