The Effects of Deferiprone and Deferasirox on the Structure and Function of β-Thalassemia Hemoglobin

Transfusional iron overload is a major cause of morbidity and mortality in thalassemia, sickle-cell disease and other chronic anemias. To overcome these problems,orally bioavailable iron chelators, deferiprone and deferasirox, were used for the treatment of patients suffering from thalassemia. The interactions between deferiprone and deferasirox with the carrier protein, beta-thalassemia hemoglobin (Hb), were investigated using fluorescence, circular dichroism (CD) and UV-visible measurements at physiological condition. Strong fluorescence quenching on interactions of the above drugs with beta-thalassemia Hb were observed. Fluorescence quenching data of thalassemia Hb in the presence of deferasirox have shown greater affinity of binding. The number of binding sites to Hb for deferasirox was found to be more relative to those of the deferiprone. The effects of these drugs on the oxygen affinity of the thalassemia Hb were studied by spectroscopic methods using sodium dithionite. Results indicated that deferiprone reduces oxygen affinity (increases oxygen releasing ability) of Hb, while in the presence of deferasirox, oxygen affinity of Hb has significantly increased by dose-dependent manner. As such, deferasirox exhibited opposite effect relative to deferiprone on the function of thalassemia Hb. In clinical dose of deferiprone, CD results showed that, the alpha-helical content of thalassemia Hb significantly increased. By use of the clinical dose of deferasirox, however, a decrease in alpha-helical content of protein was observed, which resulted in decreasing stability of thalassemia Hb. Our study showed that reduction in stability of thalassemia Hb in the presence of deferasirox induced higher conformational changes in protein.

Molecular docking further confirmed the binding site of PEG-400 in alpha-crystallin.

Heme-iron ligand (M80-Fe) in cytochrome c is destabilizing: combined in vitro and in silico approaches to monitor changes in structure, stability and dynamics of the protein on mutation

Summary: The structure, stability, and dynamics properties of horse cytochrome c and its M80G mutant have been investigated. The effect of the M80G mutation on the protein's structure and stability was examined, and the dynamic behavior of the WT cyt c and its M80G mutant was measured. The results showed that the mutation caused partial loss of structure but slightly increased overall stability, and the mutant had increased stability and flexibility compared to the wild-type.

Relationship between the Structure and Chaperone Activity of Human αA-Crystallin after Its Modification with Diabetes-Associated Oxidative Agents and Protective Role of Antioxidant Compounds

Summary: This study evaluated the impact of peroxynitrite (PON) and methylglyoxal (MGO) on alpha A-crystallin (alpha A-Cry) and found that the reaction of these compounds led to the formation of new chromophores, changes in protein structure, reduction in oligomer size, and increased chaperone activity of alpha A-Cry. Ascorbic acid and glutathione were able to reverse these effects. The results suggest that the lens antioxidant defense system, particularly glutathione, can provide strong protection against diabetic cataract.

Insulin fibrillation: Strategies for inhibition

A new nano-delivery system for cisplatin using green-synthesized iron oxide nanoparticles

Summary: This study aims to develop a new drug delivery system using green synthetic and modified iron oxide nanoparticles (IONPs) combined with the methanolic extract of Artemisia Persica loaded with cisplatin. The synthesized IONPs showed no cytotoxicity on normal cells but exhibited significant cytotoxicity on cancer cells. Additionally, the interaction between the nanoparticles and DNA did not cause any structural changes, indicating good biocompatibility. The IONP complexed cisplatin holds promise as a potential candidate in overcoming challenges of conventional chemotherapy for breast cancer treatment.

Artificial metalloenzyme with peroxidase-like activity based on periodic mesoporous organosilica with ionic-liquid framework

Summary: Artificial enzymes resembling natural enzymes were developed using a rational design approach to simulate the coordination sphere around the iron active center. Periodic mesoporous organosilica (PMO) materials were used as supports due to their unique properties, and a new peroxidase-like nanozyme was fabricated. The biomimetic pathways used in this study showed potential for the development of nanozymes with excellent thermal stability and long-term storage stability.

Effects of deep eutectic solvents on the activity and stability of enzymes

Summary: Since their introduction in 2003, deep eutectic solvents (DESs) have gained attention in various technological fields, especially biocatalysis, due to their unique properties such as low melting points, safety, low cost of raw materials, availability, sustainability, and biodegradability. Recent research has shown that enzymes can maintain and even improve their activity and stability when used in DES-based media. Therefore, DESs have emerged as promising alternatives to conventional organic solvents. This study reviews the effects of different DESs on the function and stability of various enzymes.

Exploring the Potential of ω3 Derivatives as Tyrosinase Inhibitors: A Comprehensive Study Combining Experimental, Computational, and Biological Approaches

Summary: In this study, the inhibitory ability and mechanism of omega 3-nicotinic acid (omega 3-NA) and omega 3-Picolinic acid (omega 3-PA) complexes on mushroom tyrosinase (MT) activity were investigated. The complexes showed significant inhibition on MT activity, with K-i values of 5.2 mM and 5.1 mM for omega 3-NA and omega 3-PA, respectively. Fluorescence quenching experiments confirmed the interaction between omega 3-NA, omega 3-PA, and MT. Molecular docking and dynamics simulations revealed that hydrogen bonding with Trp136, His263, and Val299 residues is involved in the formation of ligand-MT complexes. These findings suggest that omega 3-NA and omega 3-PA have the potential to be used as MT inhibitors for treating hyperpigmentation.

Study of catalase reversibility during multiple injections of H2O2 using online measurement by FFT continuous cyclic voltammetry

Summary: This study introduces a new method, FFTCCV, for determining the reversibility of catalase activation upon injection of different concentrations of hydrogen peroxide (H2O2). The results show that catalase can rapidly decompose H2O2 at concentrations below 40 mM, but its activity gradually decreases at higher concentrations. The irreversibility of catalase is triggered at a concentration of 80 mM, and the catalase activity can be recovered at lower concentrations of H2O2 after multiple injections.

Protein binding and anticancer activity of two newly synthesized Schiff base platinum (II) complexes: A theoretical and experimental study

Summary: The present study investigated the interaction of new Schiff base-platinum complexes with human serum albumin and their toxicity effects on cancer cell lines. Fluorescence and circular dichroism spectroscopy revealed the quenching mechanisms and structural changes of the albumin caused by the complexes. Molecular docking and molecular dynamics simulations identified the main interactions between the complexes and albumin. The IC50 values of the complexes on the cancer cell lines demonstrated their anti-cancer activities.
