Infections with protozoa parasites are associated with high burdens of morbidity and mortality across the developing world. Despite extensive efforts to control the transmission of these parasites, the spread of populations resistant to drugs and the lack of effective vaccines against them contribute to their persistence as major public health problems. Parasites should perform a strict control on the expression of genes involved in their pathogenicity, differentiation, immune evasion, or drug resistance, and the comprehension of the mechanisms implicated in that control could help to develop novel therapeutic strategies. However, until now these mechanisms are poorly understood in protozoa. Recent investigations into gene expression in protozoa parasites suggest that they possess many of the canonical machineries employed by higher eukaryotes for the control of gene expression at transcriptional, posttranscriptional, and epigenetic levels, but they also contain exclusive mechanisms. Here, we review the current understanding about the regulation of gene expression in Plasmodium sp., Trypanosomatids, Entamoeba histolytica and Trichomonas vaginalis.

Protein arginine methyltransferases in protozoan parasites

Mario Alberto Rodriguez

Summary: Arginine methylation is a post-translational modification that plays a significant role in protozoan parasites, particularly in pathogenicity-related events. Several PRMTs have been identified in these parasites and are involved in cell growth, stress response, stage transitions, and virulence.

An Unusual U2AF2 Inhibits Splicing and Attenuates the Virulence of the Human Protozoan Parasite Entamoeba histolytica

Summary: E. histolytica is a pathogen causing intestinal amebiasis and liver abscesses, and metronidazole is the main treatment drug. However, metronidazole-resistant strains have been reported recently, posing challenges for amebiasis eradication. RNA metabolism-related genes were found to be associated with the virulence of the parasite, and the 84KQ domain plays a critical role in splicing inhibition and intron retention. Surprisingly, the deletion of the 84KQ domain increased splicing and enhanced the virulence of the parasite.

Lysine Methyltransferase EhPKMT2 Is Involved in the In Vitro Virulence of Entamoeba histolytica

Susana Munguia-Robledo, Esther Orozco, Guillermina Garcia-Rivera, Jeni Bolanos, Jesus Valdes, Elisa Azuara-Liceaga, Mario Alberto Rodriguez

Summary: Lysine methylation, catalyzed by PKMTs, is involved in epigenetics and signaling pathways and may participate in virulence of protozoa parasites. This study found that EhPKMT2 is involved in cellular events such as phagocytosis and heat shock and has effects on cell growth, migration, and cytopathic effect, suggesting it as a potential therapeutic target for amebiasis.
