Summary: Entamoeba histolytica, a disease-causing parasitic amoeba, affects an estimated 50 million people worldwide and can lead to serious symptoms, including colitis and dysentery. Current drugs for this parasite have limitations, such as adverse effects and potential resistance. In this study, a compound library was screened and a highly potent inhibitor, JNJ001, was identified, showing better activity than the current approved treatment. Further experiments confirmed the compound's effectiveness and its inhibition of related organisms. This discovery may lead to improved therapy against this parasite.

Summary: Protozoan parasites evade host defenses by hijacking host cellular signaling pathways and transcription factors, with the NF-kappa B pathway being a key target. These parasites manipulate the NF-kappa B pathway to thwart immune responses and ensure survival in the host.

Summary: HSPC117/RtcB, a 3'-phosphate tRNA ligase, is involved in tRNA splicing and maturation, as well as mRNA splicing of certain protein-coding genes. In Entamoeba histolytica, EhRtcB1 is nuclear-localized while EhRtcB2 is cytosolic, with EhRtcB1 playing a predominant role in tRNA processing. Both EhRtcB1 and EhRtcB2 exhibit RNA ligation activity in vitro. The study demonstrates conservation of tRNA splicing and functional diversification of RtcBs in this lineage.

Summary: The protein Igl1 of Entamoeba histolytica has been found to exhibit hemolytic and cytotoxic activities. The amino acid regions 787-846, 968-1028, and 1029-1088 of Igl1 are responsible for these activities. This is the first report of a protozoan protein with hemolytic and cytotoxic activities in multiple domains.

Summary: The research focused on analyzing the genome and structure of a specific transcription factor in Entamoeba histolytica, showing differences between Myb-SHAQKYF proteins and proteins from other families. The study suggests a potential role for these proteins in the life cycle of E. histolytica.

Summary: This study characterizes, for the first time, the components of the RNA-induced silencing complex (RISC) in Entamoeba, revealing multiple functional protein components and novel nuclear assembly protein-interacting partners. The research further enhances our understanding of the molecular regulatory mechanisms of RNAi in this parasite.

Summary: This study reports the identification of a single functional Orc1/Cdc6 protein in the protozoan parasite Entamoeba histolytica and provides insights into its structural and functional characteristics using in silico analysis. The study reveals that E. histolytica possesses a less complex pre-replication machinery compared to higher eukaryotes and shares similarities with archaea. The findings lay the groundwork for further investigations into origin recognition methods and cell cycle anomalies in this enigmatic parasite.

Summary: DeepFinder is a deep learning-based tool for identifying macromolecules in cellular cryo-electron tomograms, performing with accuracy comparable to expert-supervised ground truth annotations. By using artificial neural networks, DeepFinder can simultaneously localize multiple classes of macromolecules, achieving precise localization on cellular cryo-ET data.

Summary: This study discovered a previously unknown hydrogenase in the cytosol of T. vaginalis, which can use cytochrome b5 as an electron acceptor. Trichomonads with overexpressed cytosolic hydrogenase showed decreased hydrogen excretion and increased excretion of methylated alcohols, suggesting that the cytosolic hydrogenase uses hydrogen gas as a source of reducing power for cytoplasmic reactions.

Summary: This study identified 34 GiHDMT genes in Glycyrrhiza inflata Bat. and found high conservation in gene structure and protein motifs within each group of GiHDMT genes. Various abiotic stress-related elements were detected in the GiHDMT promoters. GiHDMTs exhibited differential expression patterns across different tissues and changes in transcription level under abiotic stress. Histone methylation levels mediated by GiHDMT genes were also found to change with the treatment of NaCl and Na2CO3.

Summary: The study used PCR to identify the frequency of Entamoeba species in fecal samples from children under 5 years of age with diarrhea in Maracaibo, Venezuela. Among the samples tested, 48% of children had intestinal parasites, with E. dispar being the predominant species. However, all cases of E. histolytica infection were found in children with diarrhea, and the first case of E. moshkovskii was detected in the region.

Summary: The VHH2-Fc-VHH2 molecule, formed by fusing four VHH with one Fc, demonstrates superior quantitative antigen recognition compared to Herceptin and VHH2-Fc, as well as higher internalization capacity via the HER2 receptor. It could be a promising addition to the therapeutic arsenal with diverse applications from diagnostics to therapy.

Summary: We conducted a retrospective study on hospitalized patients in Mexico who tested positive for SARS-CoV-2 between June 2020 and January 2022. The study analyzed medical records, demographic data, comorbidities, symptoms, lab findings, and sequencing data. The findings showed differences in patient characteristics, comorbidities, and hospital stays between different waves of the pandemic. The study also revealed the presence of multiple SARS-CoV-2 clades.
