Summary: This study investigates the anti-cervical cancer activity of lipophilic grape seed proanthocyanidin (LGSP). It also effectively suppressed the growth of a HeLa xenograft tumor in a zebrafish model.

Summary: Toxin-antitoxin systems were characterized in insect cells, revealing high toxicity of the toxins and limited toxicity neutralization by the antitoxins. Both toxins induced apoptosis and necrosis in the cells, while the antitoxins partially neutralized their toxicity in a dose-dependent manner. The MazF-MazE system showed promising potential for application in insects, such as for developing new sterile insect technique (SIT).

Summary: This study evaluated the impact of rhein, an anthraquinone compound found in Chinese medicine, on the lysosomal compartment and its indirect influence on cell apoptosis. The results showed that high concentrations of rhein caused oxidative stress and changes in lysosomal membrane permeability, leading to cell apoptosis. In addition, the use of an autophagy inhibitor, chloroquine, enhanced the cytotoxicity of rhein.

Summary: This study demonstrates that the NLRP1 inflammasome detects and activates the exotoxin A released by Pseudomonas aeruginosa during chronic infection, leading to epithelial damage. The activation process is similar to other ribotoxins and is regulated by the kinase ZAKa. Cells from cystic fibrosis patients are more sensitive to ribotoxic stress and this can be inhibited by using ZAKa inhibitors.

Summary: Kashin-Beck disease (KBD) primarily affects adolescents' growth plates, influenced by both genetic and gene-environmental risk factors. This study demonstrated that HT-2 toxin is an environmental risk factor of KBD, with KBD hiPSC chondrocytes showing stronger responses to HT-2 toxin, leading to damaged organelle ultrastructure, increased G1 phase cells, and late apoptosis.

Summary: This study investigated the effect and underlying molecular mechanism of lysophosphatidic acid (LPA) on doxorubicin hydrochloride (DOX)-induced apoptosis in cervical cancer cells. The results showed that LPA could reduce cell death and chromatin aggregation caused by DOX, and downregulate the expression of caspase-3 at both mRNA and protein levels. Furthermore, LPA could prevent apoptosis in DOX-induced HeLa cells and reduce the levels of intracellular reactive oxygen species (ROS). These findings suggest that LPA may protect cervical cancer cells from DOX-induced apoptosis and could be a potential therapeutic target for the treatment of this cancer.

Summary: This study investigates the effects of a synthetic CAIX inhibitor on HeLa cancer cells. The compound I was found to inhibit cell proliferation, induce apoptosis, and affect the expression of genes and proteins related to apoptotic pathways. CAIX inhibition also caused changes in pH balance, disruption in organelle integrity, and increased intracellular reactive oxygen levels. The findings suggest that CAIX inhibition has potential in cancer treatment, and compound I could be a promising therapeutic strategy for aggressive tumors.

Summary: This study investigated the variations in triterpenoid contents and growth inhibition on HeLa cells among different Cyclocarya paliurus populations. The antiproliferation effects of leaf samples were significantly affected by pentacyclic triterpenoids, with hederagenin exhibiting the best anticancer activity. RNA sequencing identified key genes involved in apoptosis induction by cyclocaric acid B and hederagenin.

Summary: Natural products have diverse structures and novelty, leading to unique physiological activities and new action mechanisms, making them important sources for innovative drugs and lead compounds. This study demonstrates that Santamarine targets TrxR to weaken cellular antioxidative function, inducing tumor cell oxidative stress-mediated apoptosis. TrxR plays a key role in the physiological effect of Santamarine on cells, revealing a previously unrecognized mechanism of antitumor activity.

Summary: The inhibition of thioredoxin reductase (TrxR) is vital for the development of antineoplastic drugs and radiosensitizers. In this study, it is found that micheliolide (MCL), derived from Michelia, not only induces oxidative stress-mediated apoptosis in HeLa cells but also sensitizes ionizing radiation (IR) treatment by targeting TrxR. Mechanistic studies reveal that MCL covalently binds to TrxR at position 498 to inhibit its biological function. Knockdown of TrxR enhances the cytotoxicity and radiation response induced by MCL, supporting the potential of MCL as a prospective radiosensitizer and cancer chemotherapeutic agent.

Summary: Ruthenium polypyridine complexes containing 5-fluorouracil derivatives as ligands were synthesized and their anticancer activity was studied. Results showed that one of the complexes, Ru3, exhibited the best anticancer activity among the compounds tested, with selective activity. Mechanism studies revealed that Ru3 could specifically target lysosomes, induce cell apoptosis, arrest cell cycle, increase ROS level, and damage DNA, leading to cell death through the synergy of inducing apoptosis and autophagy.
